前列腺特异性抗原(PSA)是含有237个氨基酸的26kDa单链糖蛋白,由定位于人19号染色体的激肽释放酶基因编码。它由人前列腺组织特异性表达,并且是一种在精液中表达的高度丰富的蛋白水解酶。
生化/生理作用
前列腺特异性抗原(PSA)是由前列腺组织特异性产生的由雄激素调节的丝氨酸蛋白酶,并且分泌至组织腔内。精液中的PSA可裂解精液凝固物中存在的精液凝集素。前列腺癌(PCa)患者的血清中总PSA水平升高。 PSA可作为PCa的潜在生物标志物。 血清蛋白酶抑制剂如α 1-抗胰凝乳蛋白酶(α 1-ACT)、α 2-巨球蛋白(α 2-M)和其他急性期蛋白,可与循环PSA形成不可逆复合物并阻碍PSA的蛋白水解活性。
