NS8593 盐酸盐抑制小电导钙激活钾 (SKCa) 通道。
化合物 (R)-N-(苯并咪唑-2-基)-1,2,3,4-四氢-1-萘胺 (NS8593) 可逆性抑制重组 SK3 介导的电流(人和大鼠 SK3),效价约为 100 nM。NS8593 不抑制 125 I-阿帕明结合。NS8593 通过将 Ca 2 + 的激活曲线右移降低钙敏感性,仅轻微影响最大 Ca 2 + 激活的 SK 电流。NS8593 抑制所有 SK1-3 亚型 Ca 2 + 依赖性(K d = 0.42、0.60 和 0.73μM,分别为 0.5μM Ca 2 + 。NS8593 不影响中间电导和大电导的钙激活钾通道(分别为 hIK 和 hBK 通道)。NS8593 介导的抑制作用在高浓度阳性调节剂 NS309 存在时被阻止。
