應用

HMGR活性试剂盒可用于测定HMG-CoA还原酶活性。该试剂盒的主要功能是筛选HMG-CoA还原酶的纯化后催化亚基的不同抑制因子和激活因子,这在临床治疗中具有重要意义。该检测方法针对340nm处的吸光度下降进行光谱测量,而后者恰恰反映了存在HMG-CoA底物时HMGR催化亚基对NADPH的氧化水平。

生化/生理作用

HMGR(3-羟基-3-甲基戊二酰-CoA还原酶)是位于内质网上的一种跨膜糖蛋白。 这种酶可以催化HMG-CoA的四电子还原反应,生成辅酶A(CoA)和甲羟戊酸。这是固醇生物合成中的限速步骤。 为了维持衍生的甲基戊酸酯的浓度,可以通过合成、降解、磷酸化等方式控制HMGR的活性。除了HMGR的生理调控之外,人的HMGR酶可成功通过药物来靶定调控,可用于高血清胆固醇的临床治疗。 控制血清胆固醇体水平具有非常重要的治疗作用,这是因为血胆脂醇过多往往会引起动脉硬化病症加剧,进而引起心血管疾病,这可能会引起冠心病和中风。目前已有证据表明,胆固醇体内平衡受到干扰会引起慢性炎症恶化。
