Description générale

Research area: Cell CycleXanthineoxidase (XO) is a homodimer. It is made up of two sub-units each made up of onemolybdenum and two iron-sulphur (2Fe-2S) centers.[1]

Application

Xanthine Oxidase from bovine milk has been used in superoxide dismutaseactivity assay.[2][3]Xanthine oxidase may be suitable for producing amperometric biosensors and designing-flavonoid-type inhibitors against XO for treating inflammatory diseases, gout, and hyperuricemia. It may also be used in the synthesis of superoxide because of aldehyde oxidation.

Actions biochimiques/physiologiques

Although xanthine oxidase is found widely distributed in multiple tissues, it is highly expressed in endothelial and epithelial cells. Its expression was also increased in infected tissues where it was shown to have an anti-microbial effect due to its ability to generate reactive oxygen species. [4]Xanthineoxidase plays an important role in catalyzing the hydroxylation ofhypoxanthine to Xanthine. Later, xanthine is converted to uric acid isexcreted by the kidneys.[1]
Xanthine oxidase is a molybdenum-containing enzyme that is found in the cytosol, and may be strongly inhibited by flavonoids. It plays a vital role in the metabolism of some drugs, as well as purines and pyrimidines. It is also known to be a biological source of reactive oxygen species.

Définition de l'unité

One unit will convert 1.0 μmole of xanthine to uric acid per min at pH 7.5 at 25 °C. Approx. 50% of the activity is obtained with hypoxanthine as substrate.

Questions

  1. Hello, Is there a way to find out how much 2.3M (NH4)2SO4 was used to suspend 10 Units of Xanthine Oxidase? Specifically, what is the ratio of units Xanthine Oxidase (or mg) to volume needed to create this suspension using lyophilized powder? Thank you!

    1 réponse
    1. The volume of a given package size varies from lot to lot. The minimum activity of this product is 0.1 units per milligram of protein. The minimum protein concentration is 5.0 mg/mL. As an example, a lot reporting an activity of 0.3 units per milligram protein at a concentration of 13.8 mg protein per milliliter, will have a volume of 2.3 mL for the 10 Unit package size. This is determined by using the following calculation:
      10U / (0.3U/mg) = 30mg / (13.8mg/ml) = 2.3 ml volume

The lot-specific volume may be determined by data provided in the Certificate of Analysis. Please see the link below to review a sample or lot-specific Certificate:
