Pepsin is a peptidase used to digest proteins and is commonly used in the preparation of Fab fragments from antibodies. Pepsin, from porcine gastric mucosa, has been used to hydrolyze dry cervical samples in mice. Product P7125 is provided in powder form. Product P7125 has been used to denature DNA from kidney cells[1] and to digest pathology samples from anal canal carcinomas (ACC) biopsies prior to EGFR staining[2].
The enzyme from Sigma has been used to obtain total vitamin B12 content in food products prior using immunoaffinity columns[3] It has also been used to digest minced soft tissue of snails prior to the isolation of the third stage larvae (L3).[4]

Biochem/physiol Actions

It does not cleave at valine, alanine, or glycine linkages. Z-L-tyrosyl-L-phenylalanine, Z-L-glutamyl-L-tyrosine, or Z-L-methionyl-L-tyrosine may be used as substrates for pepsin digestion. Pepsin is inhibited by several phenylalanine-containing peptides.[5]

Pepsin hydrolyzes peptide bonds, not amide or ester linkages. Pepsin cleaves peptides with an aromatic acid on either side of the peptide bond. Sulfur-containing amino acids increase susceptibility to hydrolysis when they are close to the peptide bond. Pepsin preferentially cleaves at the carboxyl side of phenylalanine and leucine and at the carboxyl side of glutamic acid residues. Cleaves Phe-Val, Gln-His, Glu-Ala, Ala-Leu, Leu-Tyr, Tyr-Leu, Gly-Phe, Phe-Phe and Phe-Tyr bonds in the β chain of insulin
Pepsin is the major proteolytic enzyme produced in the stomach. It digests proteins through the cleavage of interior peptide linkages.

pH óptimo es 2-4. Activa en urea 4 M y HCl de guanidina 3 M. Estable a 60 ºC. La pepsina se desactiva de manera irreversible a pH 8,0 - 8,5.

Other Notes

One unit will produce a ΔA280 of 0.001 per min at pH 2.0 at 37 °C, measured as TCA-soluble products using hemoglobin as substrate. (Final volume = 16 mL. Light path = 1 cm.)

  1. How can I calculate the amount of pepsin P7125 needed to prepare a dissolution medium containing 750,000 units/L? Do I need to consider the reported protein percentage and the units/mg of protein to obtain the correct amount?

    1. The percentage of protein and units per mg of protein are reported in the batch-specific Certificate of Analysis. Multiplying these two values yields units per mg of solid. This value should be used to calculate activity when the powder is weighed out. For example, if the protein percentage is 10.4% and the units per mg is 1358 (lot 0000509386), the calculation would be as follows:
      10.4% × 1358 = 141.232 units/mg solid
      To prepare a dissolution medium containing 750,000 units/L, the calculation would be:
      750,000 / 141.232 ≈ 5310.41 mg
      Thus, approximately 5.3 grams of powder per liter is required.

  2. How can I find out the protein content of each lot? I want to make a 1 mg pepsin protein (not powder) / ml solution. The CoA only shows protein content is greater than1; it does not state the actual protein content for each lot.

    1. The % Protein and specific activity is published in the lot specific Certificate of Analysis (COA):

      The Specification Sheet is not lot specific, and only provides the testing criteria for each batch.

  5. Which Pepsin do you recommend for histology/antigen retrevial? According to protocol 0.2mg/ml in 0.2M HCL

    1. This product has not been specifically qualified for antigen retrieval, however, it may be able to be qualified for use by the end user. A typical protocol using pepsin for antigen retrieval suggests the following stock and working solutions:

      Pepsin Stock Solution (1% in 10mM HCl):

      Pepsin ------------------------------------- 100 mg
      10mM HCl (pH 2.0) ---------------------- 10 ml

      Mix to dissolve. Store at -20 ºC

      Pepsin Working Solution (0.5% in 5mM HCl):

      Pepsin Stock Solution (0.5%) ------------ 1 ml
      Distilled water ---------------------------- 1 ml

      Mix well.

      For antigen retrieval - cover sections with pepsin working solution and incubate for 10-20 minutes at 37 °C in humidified chamber (optimal incubation time may vary depending on tissue type and degree of fixation, and should be determined by user).
