General description

Preguntas

1–10 de 16 Preguntas
  1. How can I determine the shelf life / expiration / retest date of this product?

    1 respuesta
    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  2. I am using this kit (19160) to measure SOD activity in bacterial extracts. How can I normalize the result to protein concentration?

    1 respuesta
    1. Normalization of SOD activity to protein concentration can be achieved using the following equation: Normalized SOD Activity = SOD Activity (units/mL) / Protein Concentration (mg/mL). This calculation provides SOD activity per milligram of protein (e.g., units/mg protein), facilitating comparisons between samples or conditions and allowing for more accurate assessments of SOD activity relative to the amount of protein present.

  3. What is the meaning of sample solution? ( I have mice blood serum, can I use it directly?). Do I need individual blank 2 for all my samples and should I directly use the serum in it?

    1 respuesta
    1. The sample solution refers to what is being assayed for inhibition rate, which in this case is serum. To measure the percentage inhibition rate, various concentrations of superoxide dismutase need to be prepared. The superoxide dismutase standard can be prepared using product S7571 at concentrations of 200 U/ml, 100 U/ml, 50 U/ml, 20 U/ml, 10 U/ml, 5 U/ml, 1 U/ml, 0.1 U/ml, 0.05 U/ml, 0.01 U/ml, and 0.001 U/ml.

Sample preparation involves preparing solutions at various concentrations, as an inhibition curve requires multiple points to generate a curve. Samples should be diluted in dilution buffer or saline. Blank 2 is prepared if the sample solution has visible color and should be prepared according to Table 1. For every sample, the corresponding Blank 2 should have the same sample composition. A visual representation of a planned dilution for 1, 1/5, 1/5^2, 1/5^3, 1/5^4, 1/5^5, 1/5^6 can be prepared as a general recommendation.

  4. How is shipping temperature determined? And how is it related to the product storage temperature?

    1 respuesta
    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers. For more information, please refer to the Storage and Transport Conditions document: https://www.sigmaaldrich.com/deepweb/assets/sigmaaldrich/marketing/global/documents/316/622/storage-transport-conditions-mk.pdf

  5. Hello, Please what is the quantity of cell sample is required for use in this kit ?

    1 respuesta
    1. The recommended number of cells is 9x10*6 for adherent lines. Leukocytes have been tested successfully at 1.2x10*7 cells. Please see the link below to review the product datasheet:

  6. Can we use that kit without having SOD? And the shiping can be done at room temperature and put in fridge after reception?

    1 respuesta
    1. The superoxide dismutase (SOD) is required for preparation of an inhibition curve. This is a SOD inhibitory assay kit. See SOD product S7571, which is suggested for use with this kit. This kit is shipped on wet ice to preserve the quality of the transit. Upon receipt, the item should be stored at 2-8°C. If the interest is in calculating SOD activity in a sample, product CS0009 may be a kit of interest. See the links below to review additional information.

  7. Hello, Please what is the quantity of tissue sample is required for use in this kit

    1 respuesta
    1. This product is not routinely tested for use with solid tissue samples. However, the following general preparation method for 100 mg of tissue is recommended:
      1. Rinse the tissue with saline solution to remove residual blood, then gently dab the tissue with paper towels before proceeding to weigh it.
      2. Add 400-900 ml of sucrose buffer (0.25 M sucrose, 10 mM Tris, 1 mM EDTA, pH 7.4) and use a Teflon homogenizer to process the sample. If needed, subject the homogenized sample to sonication on an ice bath (60W with 0.5 second intervals for 15 min).
      3. Centrifuge the homogenized sample at 10,000 g for 60 min at 4ºC then carefully transfer the resulting supernatant to a new tube for measurement.

  8. What is the SOD concentration?

    1 respuesta
    1. As per the protocol of 19160: https://www.sigmaaldrich.com/deepweb/assets/sigmaaldrich/product/documents/254/301/19160dat.pdf, the kit doesn't include SOD standard, that is to say, the SOD standard needs to be purchased separately. As shown on page 2 of the protocol, if assay monitoring is required, the SOD standard should be diluted in the range of 0.001-200 U/mL, indicating the detection range of the kit is 0.001-200 U/mL.
