Descripción general

Gdown1 (DNA-directed RNA polymerase II subunit GRINL1A; DNA-directed RNA polymerase II subunit M; glutamate receptor-like protein 1A) is a nuclear protein belonging to the GRINL1 family. It binds to the RNA Polymerase II (G) molecule (RNAPII (G)) and prevents its interaction with transactivating factors, thereby suppressing RNAPII (G)-mediated transcriptional events. Gdown1 also indirectly suppresses RNAPII (G)-transcription by blocking the binding of the elongation-enhancing factor TFIIF to the elongation complex. Gdown1 also prevents TFIIF-mediated assembly of the preinitiation complex required for recruiting RNAPII to the promoter region, by binding competitively to Pol II at the RPB1 and RPB5 subunits recognized by TFIIF. Gdown1 may also inhibit the termination factor TTF2. The effects of Gdown1 are blocked by the P-TEFb. Overall, Gdown1 regulates the transcriptional activity of RNAPII (G) at multiple promoter regions by various mechanisms. Gdown1 has been detected in brain, testis, prostate, lung, small intestine, heart, kidney, and skeletal muscle.
