Allgemeine Beschreibung

Hexokinasen, die die ATP-abhängige Phosphorylierung von Aldo- und Ketohexosen in Hexose-6-Phosphat katalysieren, katalysieren den ersten Schritt in vielen wichtigen biochemischen Pfaden. Bei Säugetiergeweben ist Glucose das vorherrschende Substrat für die Hexokinasen.[1] Es gibt vier Isozyme von Hexokinase in Säugetiergewebe (HK-I, -II, -III und -IV), die sich in ihren kinetischen und regulierenden Eigenschaften sowie bezüglich der Gewebeverteilung und Kofaktor-Anwendung voneinander unterscheiden. Die Hexokinase katalysiert den ersten Schritt der meisten relevanten Glucose-Stoffwechselwege. Veränderungen der Hexokinase-Aktivität sind mit mehreren Erkrankungen assoziiert, wie der X-verknüpften Muskeldystrophie, hämolytischen Anämie und Krebs.[2]

Anwendung

Das Kalorimetrische Hexokinase-Assay-Kit wird zur Messung der Aktivität von Hexokinase eingesetzt.[3][4][5]

Eignung

Geeignet für die Messung der Hexokinase-Aktivität, Analyse des Glucosestoffwechsels und der Zellkommunikation sowie das Screening auf Diabetesmedikamente im Serum, in tierische Geweben (Leber, Herz, Niere usw.) und Zellkulturen (adhärente oder Suspensionszellen).

Prinzip

Das Kalorimetrische Hexokinase-Assay-Kit bietet ein einfaches und direktes Verfahren für die Messung der Hexokinase-Aktivität in verschiedenen Proben. Die Hexokinase-Aktivität wird durch einen gekoppelten Enzym-Assay bestimmt, bei dem Glucose in Glucose-6-Phosphat umgewandelt wird, das wiederum durch Glucose-6-Phosphat-Dehydrogenase oxidiert wird, um NADH zu bilden. Das dabei entstehende NADH reduziert eine farblose Sonde, wodurch ein kolorimetrisches Produkt (450 nm) entsteht, das proportional zur vorhandenen Hexokinase-Aktivität ist. Eine HK-Einheit ist definiert als die Menge an Enzym, die 1,0 μmol NADH pro Minute bei pH 8,0 und Raumtemperatur generiert.

  1. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.     1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
