Das GenElute HP Plasmid Maxiprep (Endotoxinfrei) Kit bietet eine einfache und kosteneffektive Methode für die Isolierung von endotoxinfreier Plasmid-DNA aus rekombinanten E. coli-Kulturen. In dem Kit kommt ein Vakuumformat mit einer Filtersäule für die schnelle Klärung des Bakterienlysats zur Anwendung und eine Kieselgelsäule dient zum Capturing der Plasmid-DNA. Eine firmeneigene Lösung sorgt dafür, dass Plasmid-DNA an eine Bindungssäule bindet, wohingegen die Adsorption von Endotoxinen verhindert wird. Mit dieser Technologie können Anwender konstante Endotoxinkonzentrationen von weniger als 0,1 Endotoxineinheiten pro μg Plasmid-DNA erreichen.

Hochwertige endotoxinfreie DNA ist für die direkte Verwendung in den anspruchsvollsten Anwendungen bereit, einschließlich der Transfektion endotoxinempfindlicher Zellen.

Anwendung

Das GenElute™ HP Plasmid Maxiprep (Endotoxinfrei) Kit wurde zur Isolierung von Plasmiden aus Schweineleber[1] und aus kompetenten Escherichia-coli-Zellen verwendet.[2]

Leistungsmerkmale und Vorteile

  • Vom Zellpellet bis zum gereinigten Plasmid in nur 35 Minuten.

Prinzip

Eine rekombinante Übernacht-E. coli-Kultur wird durch Zentrifugation geerntet und einer modifizierten alkalischen Lyse mit SDS unterzogen. Das Lysat wird durch Filtration geklärt. Anschließend wird die Bindungslösung hinzugegeben, die für endotoxinfreie Plasmidpräparationen optimiert wurde. Die Plasmid-DNA wird dann an das Kieselgel gebunden, wohingegen die Adsorption von Endotoxinen verhindert wird. Durch zwei Waschschritte werden Verunreinigungen entfernt. Schließlich wird die gebundene DNA in endotoxinfreies Wasser eluiert. Die gewonnene Plasmid-DNA liegt überwiegend in der Supercoiled-Form vor. Der Gehalt an genomischer DNA und RNA liegt unterhalb der Konzentration, die durch Agarose-Gelelektrophore mit Ethidiumbromidfärbung nachweisbar ist.

    1. The GenElute HP Plasmid Kits are much faster than other commercially available kits. A filter syringe is used for preparing the cleared lysate, which replaces a lengthy centrifugation step. These kits also offer the option of using a vacuum or spin purification format rather than a gravity drip to bind, wash and elute the plasmid. In addition, samples are eluted in Tris buffer so there are no alcohol precipitations required.

    1. Yields can reach as high as 350 mg for a midiprep and 1.2 mg for a maxiprep. As with any protocol, lower yields can be expected with low copy plasmids.
