Thorase is an ATPase that plays a critical role in regulating the surface expression of AMPA receptors (AMPAR). Thorase thus helps mediate synaptic plasticity and learning and memory. Thorase expression is required for NMDA-stimulated AMPAR internalization and for the inhibition of GRIA1 and GRIA2 recycling back to the plasma membrane as both of these require ATP processing by Thorase. Thorase is vital for keeping in balance the molecular construction-deconstruction cycle that is required for memory formation. Thorase is one of many AAA+ ATPases that drive the assembly of proteins needed to form specialized receptors at the surfaces of synapses. These receptors are stimulated by neighboring neurons, setting up the signaling and answering connections vital to brain function. Thorase regulates the all-important complementary process of receptor disassembly at synapses, which ultimately tamps down signaling. Thorase is a postsynaptic localized protein, and also is associated with the peroxisome and cell junctions and synapses. Thorase is expressed in the brain and testis primarily.
