Specificity

Clone 1F6 is a Rabbit recombinant monoclonal antibody that specifically detects GPR146. It targets an epitope within 23 amino acids from the third cytoplasmic domain within the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 23 amino acids from the third cytoplasmic domain within the C-terminal region of human GPR146.

Application

Quality Control Testing

Evaluated by Western Blotting in mouse liver tissue lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected GPR146 in mouse liver tissue lysate.

Tested applications


Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected GPR146 in human liver and 1:10,000 dilution in rat liver tissue lysates.

Affinity Binding Assay: A representative lot of this antibody bound GPR146 with a KD of 8.0 x 10-8 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected GPR146 in LoVo cells.

Target description

Probable G-protein coupled receptor 146 (UniProt: Q96CH1; also known as G-protein coupled receptor PGR8, GPR146) is encoded by the GPR146 (also known as PGR8) gene (Gene ID: 115330) in human. GPR146 is a multi-pass membrane protein with four extracellular domains, seven transmembrane domains, and four cytoplasmic domains. It is reported to promote the activity of hepatic sterol regulatory element binding protein 2 (SREBP2) via activation of the ERK signaling pathway that regulates hepatic very low-density lipoprotein (VLDL) secretion, and subsequently reduces the circulating levels of low-density lipoprotein cholesterol (LDL-C) and triglycerides (TG) levels. Deficiency of GPR146 is shown to reduce hepatic SREBP2 activity and VLDL secretion rate. This significantly reduces plasma cholesterol levels in both wild-type and LDL receptor-deficient mice. Mice deficient in GPR146 are shown to have significantly reduced aortic atherosclerotic lesions. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
