Specificity

Clone 2E11 is a Rabbit recombinant monoclonal antibody that specifically detects Reduced folate transporter (SLC19A1). It targets an epitope within 25 amino acids from the C-terminal, cytoplasmic domain.

Immunogen

KLH-conjugated linear peptide corresponding to 25 amino acids from the C-terminal, cytoplasmic domain of human Reduced folate transporter (SLC19A1).

Application

Quality Control Testing

Evaluated by Western Blotting in Human Placenta tissue lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected SLC19A1 in Human Placenta tissue lysate.

Tested applications

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected SLC19A1 in COS-7 cells.

Affinity Binding Assay: A representative lot of this antibody bound SLC19A1 with a KD of 2.1 x 10-7 in an affinity binding assay.

Target description

Reduced folate transporter (UniProt: P41440; also known as FOLT, Intestinal folate carrier 1, IFC-1, Placental folate transporter, Reduced folate carrier protein, RFC, hRFC, Reduced folate transporter 1, RFT-1, Solute carrier family 19 member 1, hSLC19A1) is encoded by the SLC19A1 (also known as FLOT1, RFC1) gene (Gene ID: 6573) in human. SLC19A1 is a multi-pass membrane protein with seven cytoplasmic domains, twelve transmembrane domains, and six extracellular domains. It is highly expressed in the liver and placenta. It serves as a transporter that mediates the import of reduced folate and a subset of immunoreactive cyclic nucleotides. It displays high affinity for N5-methyltetrahydrofolate (Km = 2-7 mM), the predominant circulating form of folate and reduced affinity for folic acid (Km = 150-200 mM). It is also involved in the import of methotrexate. Mechanistically, it acts as an antiporter that exports organic anions to facilitate the uptake of substrates. It operates optimally at pH 7.4, and its activity and folate concentrating ability is reduced with the increase in pH. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
