Specificity

Clone 3C23 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects Formin-binding protein 1 (FNBP1). It targets an epitope within 23 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 23 amino acids from the C-terminal region of human Formin-binding protein 1 (FNBP1).

Application

Anti-FNBP1, clone 3C23 ZooMAb, Cat. No. ZRB1109, is a recombinant Rabbit monoclonal antibody that specifically targets FNBP1 and is tested for use in Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected FNBP1 in NIH3T3, PC12, and U87-MG cell lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected FNBP1 in human tonsil tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected FNBP1 in THP-1 cell line and Jurkat cell line.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Target description

Formin-binding protein 1 (UniProt: Q96RU3; also known as Formin-binding protein 17, hFBP17, FNBP1) is encoded by the FNBP1 (also known as FBP17, KIAA0554) gene (Gene ID: 23048) in human. FNBP1 is a peripheral membrane protein from the cytoplasmic side. It is highly expressed in epithelial cells of the gastrointestinal tract, respiratory, reproductive and urinary systems. It is also expressed in germ cells of the testis and all regions of the brain. It is enriched in cortical regions coincident with F-actin. FNBP1 is required to coordinate membrane tubulation with reorganization of the actin cytoskeleton during the late stage of clathrin-mediated endocytosis. It binds to lipids such as phosphatidylinositol 4,5-bisphosphate and phosphatidylserine and promotes membrane invagination and the formation of tubules. Its F-BAR domain is localized to amino acids 1-264 that binds phospholipid membrane with its concave surface. FNBP1 is reported to enhances actin polymerization via the recruitment of WASL/N-WASP, which in turn activates the Arp2/3 complex. Actin polymerization may promote the fission of membrane tubules to form endocytic vesicles. A chromosomal aberration involving FNBP1 has been reported in acute leukemias. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
