General description

Product Source: Recombinant human UbcH5c Conjugating Enzyme expressed in E. coli.
The His6-tagged fusion protein of UbcH5c is charge and support ubiquitinylation in vitro. Unlike their GST-tagged counterparts, the His6-tagged UbcH5 family members all appear to form thiol ester conjugates with ubiquitin at a similar rate under similar conditions. The His-tagged version of this enzyme is not susceptible to self-ubiquitinylation. UbcH5c is highly homologous to the S. cerevisiae UbC4/5 family of E2 enzymes, demonstrated to be essential for degradation of regulatory and abnormal proteins. UbcH5c, together with UbcH5a and UbcH5b, are the most active class of E2 enzymes in cell extracts and are associated with regulation of a number of transcription factors including p53, NF-κB, and c-Fos. Typical enzyme concentration to support conjugation in vitro is 100nM to 1μM depending upon conditions.

Preparation Note

Stable for 1 year at -80°C from date of shipment. Avoid multiple freeze/thaw cycles. After initial defrost, aliquot into individual tubes and refreeze at -80°C.

Nuclear factor kappaB1 (NF-kappaB) is a heterodimeric complex that regulates transcription of many genes involved in immune and inflammatory responses. Its 50-kDa subunit (p50) is generated by the ubiquitin-proteasome pathway from a 105-kDa precursor (p105). We have reconstituted this proteolytic

Two very closely related human E2 ubiquitin conjugating enzymes, UbfH5B and UbcH5C, have been identified. These enzymes are products of distinct genes and are 88-89% identical in amino acid sequence to the recently described human E2, UbcH5 (now designated UbcH5A)
