General description

La mezcla SYBR^® Green JumpStart^™ Taq ReadyMix^™ combina los mayores rendimientos del anticuerpo JumpStart Taq para PCR de inicio en caliente con SYBR Green y la comodidad de una disolución ReadyMix lista para usar. Esta mezcla lista para usar de SYBR Green I, ADN polimerasa Taq JumpStart, desoxinucleótidos puros al 99 % y tampón de reacción se suministra en una concentración x2 para facilitar su uso. Sólo tiene que añadir 25 μl de la mezcla ×2 a la plantilla de ADN, los cebadores y el agua. El anticuerpo JumpStart Taq inactiva la ADN polimerasa a temperatura ambiente. Cuando la temperatura se eleva por encima de 70 °C en la primera etapa de desnaturalización del proceso de ciclado, el complejo se disocia y la polimerasa se vuelve completamente activa. La ADN polimerasa Taq JumpStart evita la amplificación inespecífica lo que produce valores de C_T más precisos.

SYBR^® Green JumpStart^™ Taq ReadyMix^™ has been used:

• to prepare the real-time PCR mixture
• in the quantitative analysis of gene expression by real time, quantitative polymerase chain reaction (qPCR)[1]
• to determine the mRNA expression of nuclear factor erythroid 2 (NFE2)-related factor 2 (Nrf2) gene by qPCR[2]

SYBR^® Green JumpStart^™ Taq ReadyMix^™ has been used:

• During RNA extraction, reverse transcription and Quantitative PCR[3]
• During analysis of leptin and leptin receptor expression. (PCR and melting curve analyses were performed)[4]
• For tissue RNA Isolation. (PCR and melting curve analyses were performed)[5]
• in real-time PCR (RT-PCR) of reverse-transcribed cDNA template[6][7]
• For qPCR amplifications using SYBR

Biochem/physiol Actions

La mezcla maestra SYBR Green JumpStart Taq ReadyMix se recomienda para experimentos de amplificación en tiempo real de un solo producto y también puede utilizarse para la optimización de la PCR antes de la fabricación de las sondas marcadas con fluorescencia. No se recomienda utilizar sondas marcadas con fluorescencia con el colorante SYBR Green I.

El SYBR Green I, un colorante fluorescente de unión al ADN de uso habitual, se une a todo el ADN bicatenario y la detección se controla midiendo el aumento de la florescencia durante el ciclo. El SYBR Green I tiene un máximo de excitación en 494 nm y de emisión en 521 nm.
