Application

N-Acetyl-L-cysteine has been used as a component of intestinal basal medium for the culture of mouse intestinal stem cells and also as a component of expansion medium.[1][2]

Biochem/physiol Actions

Antioxidant and mucolytic agent. Increases cellular pools of free radical scavengers. Reported to prevent apoptosis in neuronal cells but induce apoptosis in smooth muscle cells. Inhibits HIV replication. May serve as a substrate for microsomal glutathione transferase.

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.   2. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

    1. This product is not supplied sterile. However, if the water used is ultrapure, cell culture grade, and prepared in a sterile environment, there is no need to filter the solution.

  4. Can aliquots be freeze-thawed more than once?

    1. Product A9165 recommended storage is 2-8°C, Regrettably, information about stability after the material is reconstituted is unavailable. Generally speaking, avoid repeated freeze-thawed.

  5. Can aliquots be stored long term at -80 Celsius?

    1. As mentioned in the previous Q&A, it is recommended to store the aliquoted solutions at -20 °C for up to 1 month. Stability testing at -80°C has not been performed. It is not known if lower storage temperatures will extend the stability of the solution.

  6. How can I make 1M N-acyteyl cysteine solution? Should I dilute in PBS or distilled water?

    1. This product can be dissolved in H2O up to 100 mg/mL (with heating) . PBS can be used as well. Aliquot and store solutions at -20 deg. C up to 1 month.

    1. As listed on the product detail page, the molecular weight is determined as 163.19

  8. My media turned yellow when I added N-acetyl cysteine (NAC), Product A9165.  Why?

    1 answer
    1. The pH of a 1% solution is 2.0-2.8.  You may need to adjust the pH of the media with NaOH.  Or, prior to addition to the media, you can initially dilute the NAC with PBS, and then add it to the culture media.

  9. What concentration of Product A9165, N-Acetyl-L-cysteine (NAC), should I use for cell culture?

    1. Cell culture testing is performed at 0.82 g/L.  The exact concentration will depend upon your application.
