General description

Application

La poli-L-ornitina è stata usata con successo per l′attacco e lo studio della differenziazione e della crescita delle cellule neuronali. Raccomandata come substrato per colture cellulari utilizzando 0,5 mL di una soluzione 0,1 mg/mL per rivestire 25 cm2.
La soluzione di poli-L-ornitina è stata usata per indurre la differenziazione.

  1. will it be effective to reuse poly-L-ornithine (PLO), either in diluted or undiluted form? Following a coating period of 1 hour or overnight, can the residual PLO solution be collected, stored at 4 °C, and subsequently reused for additional coatings?

    1. The suggested coating protocol involves removing excess solution by aspiration and rinsing the vessel with sterile water. Collecting residual poly-L-ornithine (PLO) solution and storing it at 4 °C for reuse has not been tested internally. External sources indicate that the PLO solution can be reused after coating, and this practice is common in many laboratories. When reusing the PLO solution, it is essential to utilize a sterile hood and aseptic techniques to prevent contamination.

    1. This item is stored refrigerated at 2-8°C. However, the product is shipped at ambient temperature. Shipping routes are configured to minimize transit time.
      Refrigerated products that ship at ambient temperature are considered stable up to 2 weeks for transit time.

  3. CAt#P4957 do I just coat with the stock solution? at what conditions? RT or 37C or other? For how long?

    1. This product is a 0.01% or 100 ug/mL solution. The recommended surface coverage for this product is 4 μg/cm2. Provided the surface area of the vessel accommodates this concentration, the product may be used undiluted. A T-25 Tissue Culture flask has a surface area of 25 cm2.

      In this case. 1 mL of the undiluted product would be sufficient to coat the entire culture surface. Conversely, a 96 well flat bottom plate will have a culture area of approximately 0.150cm2 requiring approximately 0.6 ug to coat the surface. In this example, it may be more practical to dilute to a practical volume. Surfaces may be coated at room temperature. Incubate at least 5 minutes before removing excess solution. Coated vessels may be sterilized by UV and stored in a sealed container for up to 1 year. Avoid using coated vessels or slides that have any yellowing or changes in the surface appearance as this is an indication of degradation.

  4. Will poly-L-ornithine still be effective if it is incubated for 2 hours at 37°C, as done with other substrates, instead of being coated at room temperature overnight as indicated?

    1. The optimal coating conditions, including the required incubation time and temperature, are application-dependent and should be determined empirically. For reference, this product has been reported to coat well at 37°C with just 30 minutes of incubation time in a publication that may be of interest (Nature. 2019 Aug;572(7770):543-548. doi: 10.1038/s41586-019-1464-0. Epub 2019 Aug 7. Pol II phosphorylation regulates a switch between transcriptional and splicing condensates, PMID: 31391587).
