Descrizione generale

Choline is an essential nutrient, commonly grouped with the B complex vitamins, that plays key roles in many biological processes. Choline is a precursor for the synthesis of acetylcholine, a critical neurotransmitter in both the peripheral and central nervous systems.

Applicazioni

Choline/Acetylcholine Quantification Kit has been used to quantify the amounts of choline and acetylcholine in samples.[1][2]

Compatibilità

Suitable for use with cell and tissue culture supernatants as well as serum, plasma, urine, and other biological fluids.

Principio

In this assay, choline concentration is determined by a coupled enzyme reaction, which results in a colorimetric (570 nm)/fluorometric (λex = 535/λem = 587 nm) product, proportional to the choline present. Acetylcholine levels can be determined by adding acetylcholinesterase to the reaction, which hydrolyzes acetylcholine to choline and acetate.

  1. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  2. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.   3. Does Choline/Acetylcholine Quantitation Kit (Catalog Number MAK056) have reactivity with plasma of dog or canine?

    1. This kit may be suitable with plasma from a canine. Species reactivity is not determined, however, the publications available in the PEER REVIEWED PAPERS section applied this kit to mouse, rat, squirrel, and human samples. There is no reason to suspect canine plasma would not be suitable.

  4. Do I need to prepare two seperate standard sets for free and total choline prepared with different reaction mixes?

    1. While the kit protocol recommends running samples and standards in duplicate, a second set of standards treated with acetylcholinesterase is not required to determine both free and total choline. This can be determined mathematically. See the link below to review the protocol. See page 2 for the free and total choline reaction mix set up. The calculations for Acetylcholine can be found on page 3 in the upper right column.

  5. To prepare choline standards for , fluor. detection0.05 mM standard solution is mixed it with Choline assay buffer to bring the volume to 50 uL. 50 ul of reaction mix will be added to standard wells to bring final volume of 100 ul. Is this correct?

    1. Yes, the final Choline Standard solution is diluted with the Choline Assay buffer to a volume of 50 ul. The 100 ul final volume is comprised of 50 ul of the reaction mix and 50 ul of the standard or sample to be tested.
