Descrizione generale

GTP functions as a carrier of phosphates and pyrophosphates involved in channeling chemical energy into specific biosynthetic pathways. It also serves as an energy-rich precursor of mononucleotide units in the enzymatic biosynthesis of DNA and RNA.[1] GTP is involved in the initiation of peptide synthesis during the binding of formylmethionyl-tRNA (F-met-tRNA) to the ribosome.[2][3] It is also involved in polypeptide chain elongation.[4] GTP is hydrolyzed by tubulin and its hydrolysis is accompanied by microtubule assembly.[5]
GTP is supplied as the disodium salt in crystal form.

Applicazioni

GTP, disodium salt, is required as a coenzyme for protein biosynthesis in a cell-free system. It has been used in FtsZ polymerization assay[6] and in the preparation of pipette solution for the electrophysiological recordings of the whole-cell membrane currents.[7]

Nota sulla preparazione

Activator: EDTA prevents the formation of enzyme aggregates. Phosphate stimulates the NADH-dependent reaction.
Storage conditions (working solution): Aqueous solutions (pH 7.0) are stable at 2 to 8 °C or frozen for approx. one week, for reasons of hydrolysis pH should not be < 3.

If neutralized with water preparation can be stored frozen at -15 to -25 °C in small portions, one sample should thawed only once and should be used within the next one or two days (stored at 2 to 8 °C). Preparation can also be dissolved in a buffer pH 7-8 without neutralization (buffer concentration should be high enough that pH is not changed by addition of those substances); solution can be stored frozen.

Investigations on the storage temperature 2 to 8 °C compared to -15 to -25 °C showed no difference.
