Spezifität

Clone 1M9 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Isocitrate dehydrogenase [NADP] cytoplasmic (IDH1). It targets an epitope within 14 amino acids from the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 14 amino acids from the C-terminal half of human Isocitrate dehydrogenase [NADP] cytoplasmic (IDH1).

Anwendung

Quality Control Testing

Evaluated by Western Blotting in Mouse Kidney tissue lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected IDH1 in Mouse Kidney tissue lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected IDH1 in C2C12 and NIH3T3 cell lysates.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected IDH1 in NIH 3T3 cells.

Affinity Binding Assay: A representative lot of this antibody bound IDH1 with a KD of 5.9 x 10-10 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected IDH1 in mouse testis tissue sections.

Zielbeschreibung

Isocitrate dehydrogenase [NADP] cytoplasmic (UniProt: EC:1.1.1.42, IDH, Cytosolic NADP-isocitrate dehydrogenase, IDP, NADP(+)-specific ICDH, Oxalosuccinate decarboxylase) is encoded by the IDH1 (also known as PICD) gene (Gene ID: 3417) in human. A total of five isocitrate dehydrogenases have been described: three of these are NAD(+)-dependent that localize to the mitochondrial matrix, and two are NADP(+)-dependent enzymes. One of the NADP(+) enzyme is mitochondrial and the other is predominantly localized in the cytoplasm. IDH1 is a homodimeric enzyme that is localized in cytoplasm and peroxisomes and catalyzes the oxidative decarboxylation of isocitrate to 2-oxoglutarate. It has two NADP binding sites (aa 75-77 and 310-315) and plays a significant role in cytoplasmic NADPH production. Acetylation at Lys-374 is reported to significantly reduce its catalytic activity. Mutations in IDH1 gene, particularly those affecting Arg 132, have been linked to development of high-grade gliomas. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physikalische Form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Rekonstituierung

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Lagerung und Haltbarkeit

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
