Application

Alcohol Dehydrogenase from Saccharomyces cerevisiae has been used as a gel filtration molecular weight marker/ It has also been used as a component of nine protein mixture for mass spectroscopy analysis.

Biochem/physiol Actions

ADH (Alkoholdehydrogenase) ist eines der ersten Enzyme, die isoliert und aufgereinigt wurden. NAD⁺ ist sein Kofaktor. Es wurden drei Isozyme von Hefe-ADH, d. h. Hefe-Alkoholdehydrogenase 1, 2 und 3 (Yeast Alcohol Dehydrogenase, YADH) nachgewiesen. YADH-1 wird während anaerober Fermentation exprimiert, YADH-2 wird im Zytoplasma exprimiert und YADH-3 ist in den Mitochondrien lokalisiert. Ein 141kDa-Tetramer mit 4 gleichen Untereinheiten. Die aktive Stelle jeder Untereinheit enthält ein Zinkatom. Jede aktive Stelle enthält außerdem 2 reaktive Sulfhydrylgruppen und einen Histidinrest.

Isoelektrischer Punkt: 5,4–5,8

Optimaler pH-Wert: 8,6–9,0

Substrate: Hefe-ADH ist am aktivsten mit Ethanol. Ihre Aktivität nimmt mit wachsender oder abnehmender Größe des Alkohols ab. Verzweigtkettige Alkohole und sekundäre Alkohole weisen ebenfalls eine sehr niedrige Aktivität auf.

Alcohol Dehydrogenase (ADH) is an oxidoreductase and also a pyridine nucleotide-dependent dehydrogenase. It catalyzes the generation of aldehydes or ketones by reversible oxidation of alcohols. ADH in parallel also mediates the reduction of the nicotinamide adenine dinucleotide (NAD⁺) or nicotinamide adenine dinucleotide phosphate (NADP⁺). ADH from yeast is more active than mammalian ADHs.
