Spécificité

Clone 1H8 is a mouse monoclonal antibody that detects Anthrax toxin receptor 2 (ANTXR2). It targets an epitope within the N-terminal half.

Immunogène

A recombinant fragment of 178 amino acids from the N-terminal half of human ANTXR2 with an N-terminal extension of GSHMLEDPRG.

Application

Quality Control Testing

Evaluated by Western Blotting in Mouse lung tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected ANTXR2/CMG-2 in Mouse lung tissue lysate.

Tested Applications

Western Blotting Analysis: A representative lot detected ANTXR2/CMG-2 in Western Blotting applications (Chaudhary, A., et al. (2012). Cancer Cell. 21(2):212-26; Szot, C., et al. (2018). J Clin Invest. 128(7):2927-2943).

Description de la cible

Anthrax toxin receptor 2 (UniProt: P58335; also known as Capillary morphogenesis gene 2 protein, CMG-2) is encoded by the ANTRX2 (also known as CMG2) gene (Gene ID: 118429) in human. ANTRX2/CMG-2 is widely expressed, single-pass type I membrane protein that is synthesized with a signal peptide (aa 1-33), which is subsequently cleaved off to produce the mature form that contains an extracellular domain (aa 34-318), a transmembrane domain (aa 319-341), and a cytoplasmic domain (aa 342-489). It shares about 58% amino acid identity with ANTXR1. Four isoforms of ANTRX2/CMG-2 have been described that are produced by alternative splicing. It is reported to be essential for cellular interactions with laminin and the extracellular matrix. It is an important regulator of collagen VI homeostasis. It mediates the transport of collagen to lysosomes for degradation. Its loss promotes accumulation of Collagen VI leading to nodule formation. Antrx2 -/- mice are shown to display massive accumulation of collagen VI in their uteri leading to fibrosis and sterility. It binds to collagen VI via its vWA domain (aa 44-213) in a MIDAS (metal ion-dependent binding domain) manner. (Ref.: Burgi, J., et al. (2017). Nat. Commun. 8; Article 15861; Chaudhary, A., et al.
