Descrição geral
Trata-se de um pequeno agente alquilante lipofílico[1] que interage covalentemente com o ambiente nucleofílico dentro do DNA (resíduos de guanina). Devido a essa propriedade, a temozolomida é considerada um agente citotóxico. O mecanismo de ação da TMZ depende de sua capacidade de hidrolisar o DNA, resultando na degradação do DNA e destruição das células tumorais. Células de glioma maligno respondem principalmente à TMZ por meio de arresto do ciclo celular na fase G2/M ou, em casos raros, por apoptose.[1] A temozolomida é um agente de metilação de DNA e agente modificador da resistência a medicamentos; antitumoral e antiangiogênico. A temozolomida induz o bloqueio de G2/M e a apoptose por meio da adução de um grupo metil à posição O6 da guanina no DNA genômico e inativação funcional da proteína de reparo de DNA O(6)-alquilguanina DNA alquiltransferase (AGT) na via de reparo por excisão de base (BER).
Aplicação
A temozolomida tem sido utilizada para analisar mecanismos de resistência a medicamentos em linhagens celulares de glioblastoma. A temozolomida tem sido usada para induzir efeitos citotóxicos em células de glioblastoma para estudar o impacto da inibição da proteína disulfeto isomerase (PDI).[2]

How long is Temozolomide stable for when dissolved as recommended, in DMSO at 10 mg/mL and storaged at 2-8C? Also, can the solution only be stored at 2-8C or can it be stored at -20C or -80C for long term storage

Wherever possible, solutions should be prepared fresh and used on the same day. However, should stock solutions need to be prepared in advance, store in aliquots at -20°C for up to 1 month. Allow aliquots to equilibrate to room temperature prior to opening. The long term storage at other temperatures is not determined. However, the published reference below may prove helpful:
