Descrição geral

CDK9/CyclinK, active, GST tagged human comprises recombinant full-length human CDK9 and cyclinK proteins co-expressedin Sf9 insect cells with an N-terminal GST tag. Cyclin-dependent protein kinase 9 (CDK9)/CyclinK is a member of the cyclin-dependent protein kinase (CDK) family. CDK9 is a component of the multiprotein complex mitogen-activated protein kinase 7 and transcription elongation factor β (TAK/P-TEFβ).[1]

Ações bioquímicas/fisiológicas

CDKs are closely related to yeast CDk1, cdc28 and cdc2 and arean important regulators of the cell cycle.[2] CDK9 can modulate RNA polymerase II-directed transcription by phosphorylating the C-terminal domain of the largest subunit of RNA polymerase II.[3] CDK9 forms a complex with and is regulated by its regulatory subunit cyclin T or cyclin K. CDK9 also interacts with the human immune deficiency virus 1 (HIV-1) transactivating regulatory protein (Tat) protein, which suggests a possible involvement of this protein in AIDS.[1]

Cervical cancer is the fourth most frequently diagnosed and fatal gynecological cancer. 15-61% of all cases metastasize and develop chemoresistance, reducing the 5-year survival of cervical cancer patients to as low as 17%. Therefore, unraveling the mechanisms contributing to metastasis

Orderly progression through the cell cycle requires sequential activation and inactivation of cyclin-dependent kinases (cdks). This is achieved in part through the association of cdks with positive regulators called cyclins and inactivation of cyclin-cdk complexes by a rapidly growing number

The human positive transcription elongation factor P-TEFb, consisting of a CDK9/cyclin T1 heterodimer, functions as both a general and an HIV-1 Tat-specific transcription factor. P-TEFb activates transcription by phosphorylating RNA polymerase (Pol) II, leading to the formation of processive elongation
