Description générale

This product is a TCA extract of E. coli serotype O55:B5 (strain CDC 1644-70), which was purified by gel filtration and conjugated to FITC (fluorescein isothiocyanate).

Application

Les lipopolysaccharides (LPS) sont des constituants caractéristiques de la paroi cellulaire des bactéries à Gram négatif. Les LPS et leur fraction lipidique (lipide A) stimulent les cellules du système immunitaire inné via le récepteur TLR4 (Toll-Like Receptor 4), un membre de la famille des récepteurs de type Toll, qui reconnaît la signature moléculaire des principaux agents pathogènes (PAMP).
FITC-Lipopolysaccharides (FITC-LPS) may be used to study the movement/transport of LPS across tissue barriers such as the colonic epithelium and cellular binding and internalization of lipopolysaccharides with fluorescence-based assay systems.[1][2] FITC-LPS may be used to evaluate, qualify and quantify lipopolysaccharides (LPS) removal reagents and procedures.[3] FITC-LPS conjugate may be used for the detection of LPA binding proteins and motifs.[4]

Actions biochimiques/physiologiques

Lipopolysaccharides (LPS) are localized in the outer layer of the membrane and are, in noncapsulated strains, exposed on the cell surface. They contribute to the integrity of the outer membrane, and protect the cell against the action of bile salts and lipophilic antibiotics.

Notes préparatoires

The product is soluble in water (5 mg/ml), yielding a hazy faint yellow solution. Lipopolysaccharides are molecules that form micelles in every solvent. Hazy solutions are observed in water and phosphate buffered saline. Organic solvents do not give clearer solutions. Methanol yields a turbid suspension with floaters, while water yields a homogeneously hazy solution.

    1. The molecular weight of this product is not determined. However, LPS is heterogeneous and tends to form aggregates of varying sizes that may range from 1-4 million or greater. When treated with SDS and heat the molecular weight range is approximately 50 to 100 kDa. The FITC used to prepare this product has a molecular weight of 389.38 g/mol. There are approximately 3.6 ug FITC per milligram LPS in this preparation.
