Application

L′hémine est utilisée dans les applications suivantes :
  • Pour étudier des souches bactériennes et leurs conditions de croissance[1]
  • Dans des essais à court terme sur des colonies (avec un milieu de culture constitué d′hémine et d′autres éléments)[2]
  • Pour des transfections cellulaires[3]
Elle peut être utilisée dans des essais de formation de β-hématine afin d′analyser l′activité inhibitrice des composés de la Malaria Box (une liste de composés à activité antipaludique) du projet MMV (Medicines for Malaria Venture) contre la formation de β-hématine.[4]

Actions biochimiques/physiologiques

L′hémine, la version oxydée de l′hème, est un groupement prosthétique contenant du fer présent dans un vaste groupe de protéines. L′hème libre, qui peut être libérée par l′hémoglobine après hémolyse, est pro-inflammatoire et contribue à former des espèces réactives de l′oxygène dérivées du fer. Les taux d′hémine libre peuvent être régulés à la hausse dans diverses pathologies et peuvent contribuer au développement de divers troubles inflammatoires, notamment des affections vasculaires, une insuffisance rénale et des troubles à médiation immunitaire.

L′hémine peut servir à étudier l′expression et la répression de l′hème oxygénase 1 (HO-1) ainsi que l′activité de la HO-1. L′hématine est le « facteur X » nécessaire à la croissance d′Haemophilus influenzae.

If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.     1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.

    1. The product with code 51280 can be dissolved in DMSO at a concentration of 1 mM. It is also soluble in 1.4 M ammonium hydroxide at a concentration of 50 mg/ml, and this can be achieved by heating the solution at 95°C for 5 minutes, resulting in a dark brown solution. Additionally, a stock solution of 0.25% hemin in 50% triethanolamine can be prepared. CAS number 16009-13-5, which corresponds to product 51280, is soluble in 1.4 M NaOH at a concentration of 25 mg/mL, and it is practically insoluble in water.

  4. How can hemin be dissolved for cell culture experiments in macrophages? Is it necessary to couple it to serum/BSA?

    1. This product has not been tested for use in cell culture. However, a previously deleted product, H1652, has been tested. The H1652 was dissolved at 10 mg/mL in 1 M NaOH and added then added to the culture medium at 7 mg/L. This information would need to be validated for product 51280.
