A cyclic nucleotide-independent serine/threonine protein kinase composed of catalytic α-subunits and regulatory β-subunits in an α2β2- tetramer. This material is one of several forms of native CK-II. Suitable for in vitro phosphorylation of proteins and peptides, phosphopeptide synthesis, characterization of phosphorylating sites, screening of activators and inhibitors, and mechanistic studies. The following substrates have been characterized by CK-II both in vitro and in vivo: nucleolin, RNA polymerase, p53, and SV 40 large T antigen.

- has been used as a marker for CK2 subunits, to analyze CK2 expression in human retinal extract.
- to investigate the phosphorylation of the C-terminal domain of uroplakin IIIa and assess its potential regulation by CK2.
- to investigate the phosphorylation events and their impact on spectrin tetramer formation and neuronal cell function.
Actions biochimiques/physiologiques
Casein kinase II (CK2) is an active serine/threonine protein kinase that plays a pivotal role in cell growth and division. It is responsible for phosphorylating various nuclear proteins involved in DNA metabolism, transcription factors, and oncogenes, thus influencing their activity. It also regulates osteogenesis, adipogenesis, chondrogenesis, neuron differentiation, and immune response pathways. CK2 plays a crucial role in activating the Janus Kinase/signal transducer and activator of transcription (JAK/STAT) pathway, which contributes to cell survival, differentiation, and proliferation. Furthermore, CK2 is involved in activating the nuclear factor kappa B (NF-κB) pathway. It is highly expressed in different hematological malignancies, including acute lymphoblastic leukemia (ALL), acute myeloid leukemia (AML), chronic lymphocytic leukemia (CLL), and chronic myeloid leukemia (CML).
