Sodium channel protein type 2 subunit alpha (UniProt: Q99250; also known as HBSC II, Sodium channel protein brain II subunit alpha, Sodium channel protein type II subunit alpha, Voltage-gated sodium channel subunit alpha Nav1.2, SCN2A) is encoded by the SCN2A (also known as NAC2, SCN2A1, SCN2A2) gene (Gene ID: 6326) in human. Sodium channels are integral plasma membrane proteins that conduct Na+ transport across the plasma membrane. They are classified as either voltage-gated or ligand-gated based on the nature of the triggering mechanism for channel activation. SCN2A is a multi-pass membrane glycoprotein with 13 cytoplasmic domains, 16 extracellular domains, and 24 transmembrane domains, and It mediates the voltage-dependent sodium ion permeability of excitable membranes. It forms a sodium-selective channel through which Na+ ions may pass in accordance with their electrochemical gradient. Mutations in SCN2A gene are known to cause infantile epileptic encephalopathy that is characterized by infantile-onset seizures, before 12 months of age, followed by neurodevelopmental delay. Application

Quality Control Testing

Evaluated by Western Blotting in Human brain tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected SCN2A in Human brain tissue lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected SCN2A in Human hippocampus membrane preparations.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected SCN2A in K562 cells (not permeabilized).

Affinity Binding Assay: A representative lot of this antibody bound SCN2A in SCN2A Sodium Channel with a KD of 1.2 x 10-8 in an affinity binding assay.

Biochem/physiol Actions

Clone 3F19 is a ZooMAb^® Rabbit recombinant monoclonal antibody that specifically detects Sodium channel protein type 2 subunit alpha (SCN2A). It targets an epitope within 23 amino acids from the C-terminal, cytoplasmic domain.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb^® formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Preparation Note

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
