Application

Tris Acetate-EDTA buffer has been used as run buffer for the agarose gel electrophoresis of human papillomavirus DNA and transcribed RNA samples.
TAE-Laufpuffer ist der in der DNA-Agarosegel-Elektrophorese am häufigsten verwendete Puffer, wird aber auch in der nicht-denaturierenden RNA-Agarosegel-Elektrophorese eingesetzt. Doppelsträngige DNA läuft in TAE tendenziell schneller als in anderen Puffern, kann sich aber während erweiterter Elektrophorese erschöpfen. Eine Pufferzirkulation oder ein Pufferaustausch während erweiterter Elektrophorese kann die geringere Pufferungskapazität beheben. Die Verdünnung des konzentrierten TAE-Puffers ergibt einen 1× TAE-Puffer mit 40 mM Tris-Acetat und 1 mM EDTA, pH 8,3. Der 1× TAE-Puffer wird sowohl im Agarosegel als auch als Laufpuffer verwendet. Für maximale Auflösung werden angewendete Spannungen von unter 5 V/cm (Abstand zwischen den Elektroden des Geräts) empfohlen.

Preparation Note

Wird mit BioPerformance-zertifizierter Trizma-Base (T6066) und EDTA-Dinatriumsalz der molekularbiologischen Güteklasse (E5134) hergestellt. Lösungen enthalten außerdem Essigsäure (A6283), die pulverisierte Mischung enthält Trizma-Acetat (T1258).
Solution prepared with 18 megohm water

Other Notes

40 mM Tris acetate, pH approx. 8.3, containing 1 mM EDTA.

  1. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  2. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.   3. Could you provide the concentration of product T6025 and clarify if it is considered 50X, 10X, or 1X?

    1. The concentration for product T6025 is 40 mM Tris acetate with 1 mM EDTA at approximately pH 8.3. This solution, prepared with 18 megohm water and 0.2 μm filtration, is considered a 1X working solution.
