Application
Dieses Produkt von Sigma dient zum Nachweis, dass Gewebetransglutaminase (tTG) Glutenpeptide selektiv desamidiert, was zu einer stark erhöhten T-Zell-stimulierenden Aktivität führt.[1] Es wird außerdem zur Beurteilung von Immunantworten auf A-Gliadin-Peptide verwendet.[2] Darüber hinaus dient es zum Nachweis, dass tTG Gliadinpeptide, die bei Zöliakie von aus dem Darm stammenden T-Zellen erkannt werden, selektiv modifiziert.[3]
Transglutaminase wurde in einer Studie dazu verwendet, quantifizierbare Assays dahingehend zu verbessern, dass die Rolle der Transglutaminase bei Erkrankungen wie Chorea Huntington und Alzheimer vollständig charakterisiert wird. Transglutaminase wurde außerdem in einer Studie zur Entwicklung eines nicht radioaktiven Dot-Blot-Assays für Transglutaminase-Aktivität eingesetzt.
Biochem/physiol Actions
Transglutaminase aus Meerschweinchenleber besteht aus einer einzelnen Polypeptidkette mit 691 Aminosäureresten. Sie hat sechs potenzielle Gylcosylierungsstellen (Asn-X-Ser oder Asn-X-Thr), ist aber nicht glycosyliert. Die Molekularmasse beträgt ca. 76,6 kDa.[4] Sie ist calciumabhängig und hat mehrere calciumbindende Stellen. Das Enzym wird in Gegenwart von Calcium durch Iodoacetamid und N-Ethylmaleimid gehemmt.[5] Es katalysiert den Einbau niedermolekularer Amine in γ-Glutaminstellen von Proteinen. Bei Abwesenheit von niedermolekularen Aminen katalysiert es die Vernetzung von Proteinen, was zur Bildung von γ-Glutamyl-ε-Lysin-Seitenkettenpeptiden führt. Lebertransglutaminase ist ein nicht zymogenisches Enzym.[6]
