General description
Trichostatin A ist eine Verbindung, die hauptsächlich aus Hydroxamsäure besteht.
Application
Trichostatin A als fertige Lösung wird verwendet:
- als Histon-Deacetylase-Inhibitor zur Untersuchung von dessen Wirkung auf Transkriptomänderungen während der Stammzellentestung
- zur Hemmung von Histon-Deacetylase (HDAC) Klasse I, II oder III in primären hypophysären Zellkulturen und zur Untersuchung der Insulinregulierung des Gens für das endogene Wachstumshormon im Menschen (hGH)
- zur Behandlung von Zellen bei Experimenten zur HDAC-Hemmung
Biochem/physiol Actions
Trichostatin A (TSA) ist ein Streptomyces-Metabolit, der bei nanomolarer Konzentration spezifisch die Histondeacetylase in Säugetieren hemmt und die Anlagerung von stark acetylierten Histonmolekülen in Säugetierzellen bewirkt. Aus diesem Grund wird Trichostatin A zur Studie der Folgen von Histonacetylierung in vivo genutzt. Trichostatin A induziert Zelldifferenzierung, Zellzyklus-Arrest, die Umkehrung der Morphologie transformierter Zellen sowie Apoptese und kann die Transkription beeinflussen. TSA wird zur Etablierung einer Klonierungstechnik zur Erhöhung der Erfolgsquote beim Klonen von Mäusen verwendet.
