Allgemeine Beschreibung

The PIWIL3 (Piwi like RNA-mediated gene silencing 3) protein belongs to the Piwi subfamily of Argonaute proteins. The Argonaute proteins contain a Piwi Argonaute-Zwille (PAZ) domain and PIWI domain.[1][2] In humans, there are four PIWI proteins. The PIWIL3 gene is mapped to human chromosome 22q11.23.[3] The Piwi proteins are mostly localized to the germline, where they bind to PIWI-interacting RNAs (piRNAs).[1]

Biochem./physiol. Wirkung

The P-element Induced WImpy testis (Piwi) proteins are important for the biogenesis and function of small non-coding RNAs. These proteins and PIWI-interacting RNAs (piRNAs) have been implicated in epigenetic control of gene expression, transposon silencing, gene expression, and translation regulation. The Piwi proteins significantly participate in germline development and gametogenesis of various metazoan species.[2] These proteins undergo symmetrical dimethyl arginines (sDMAs) modification by protein methyltransferase 5 (PRMT5).[5] The Piwi proteins are implicated in various cancers such as colorectal, breast, bladder, ovarian and non-small lung cancer and many more. Increased PIWIL3 gene expression has been observed in gastric cancer and metastatic melanoma.[3]

Lagerung und Haltbarkeit

Store at −20 °C. For continuous use, store at 2–8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation. Working dilutions should be discarded if not used within 12 hours.
