NADPH ist ein Elektronenspender und Cofaktor vieler Redox-Enzyme, einschließlich Stickoxid-Synthetase.

β-Nicotinamidadenin-Dinukleotid-2′-Phosphat (NADP+) und reduziertes β-Nicotinamidadenin-Dinukleotid-2′-Phosphat (NADPH) sind ein Coenzym und Redoxpaar (NADP+:NADPH), das an zahlreichen enzymkatalysierten Oxidations-Reduktions-Reaktionen beteiligt ist. Das Redoxpaar NADP+/NADPH ermöglicht den Elektronentransfer in anabolen Reaktionen wie der Biosynthese von Fetten und Cholesterin und der Verlängerung von Fettacylketten. Das Redoxpaar NADP+/NADPH wird in einer Vielzahl von Antioxidationsmechanismen verwendet, wo es Schutz vor der Akkumulation reaktiver Sauerstoffspezies bietet. NADPH wird in vivo über den Pentosephosphatweg (PPP) erzeugt.
