Chemische Struktur: Peptid

Application

Nisin ist ein polyzyklisches Lantibiotikum und wird von Lactococcus lactis produziert. Es wird häufig für die Nahrungsmittelkonservierung verwendet. Außerdem wird es in Zellkulturen als Selektionsmittel zur Isolierung von gramnegativen Bakterien, Hefen und Schimmelpilzen genutzt. Nisin wird in Bioassays zur Erforschung der posttranslationalen Modifikation eingesetzt.[1]
Zur Bildung gezielter Poren in Zellmembranen, die von Lipid II vermittelt werden. Zur Erforschung der Assemblierung und Stabilität von Nisin-Lipid-II-Porenkomplexen.

Biochem/physiol Actions

Nisin bindet an Zellwandvorläufer-Lipide von Bakterien und stört die Zellwandbildung. Nisin verändert die Zellmembran, was zu einem Austreten von zytoplasmischen Bestandteilen und der Zerstörung des Protonengradienten führt.[1]
Nisin ist ein Antibiotikum mit bakterizider Wirkung. Es bindet an den Lipid-A-Abschnitt bakterieller Lipopolysaccharide und induziert die Porenbildung in den Membranen von Kortexzellen aus exzidierten Sorghumwurzeln.

Wirkungsweise: Bindet an die Zytoplasmamembran und beeinflusst ihre Durchlässigkeit.
Antimikrobielles Spektrum: Gramnegative und grampositive Bakterien (Chelator wie z. B. EDTA erforderlich).

    1. The solubility has not been tested. However, several literature sources indicate that this material is soluble in dilute acids. Solubility was quite high at low pH (57 mg/ml at pH 2) and much lower at high pH (0.25 mg/ml at pH 8 to 12). Dissolving the compound in water is not recommended. In addition, the product may also be soluble in DMSO with gentle warming and ultrasonication, up to at least 5 mg/mL. This information has not been validated.
