General description

DAPI, 4′,6-diamidino-2-phenylindole, is a blue fluorescent nucleic acid stain that fluoresces brightly upon selectively binding to the minor groove of double stranded DNA. Its selectivity for DNA and high cell permeability allows efficient staining of nuclei with little background from the cytoplasm. The excitation and emission wavelengths of DAPI-DNA complex are 360 nm and 460 nm, respectively. Most of the fluorescent dyes fade (photobleach) when exposed to excitation light. The amount of photobleaching depends on the composition of the molecule and the time of light exposure. DAPI Ready Made Solution with Antifade is a formulation that suppress photobleaching and prolong fluorescent signals.

Application

DAPI ist um ein Vielfaches empfindlicher als Ethidiumbromid zur DNA-Färbung in Agarosegelen. Es kann für sogenanntes Photo-Footprinting von DNA verwendet werden, zum Nachweis gebundener Sonden in Blotting-Anwendungen durch gezielte Visualisierung des doppelsträngigen Komplexes, als auch um Änderungen in der DNA zu untersuchen und um den DNA-Gehalt während der Apoptose mittels Durchflusszytometrie zu bestimmen. DAPI-Färbung ist eine sensitive und spezifische Nachweismethode für Mykoplasmen.
DAPI ist um ein Vielfaches sensitiver als Ethidiumbromid für die Färbung von DNA in Agarosegelen.
DAPI Ready Made Solution with Antifade is a glycerol based ready to use solution provided at 1μg/mL concentration in a dropper vial that can be applied directly to cells or tissue samples on microscope slides or coverslips. It is recommended to warm up at room temperature and mix by swirling the dropper vial before use. Store DAPI Ready Made Solution with antifade at -20 C protected from light.

Biochem/physiol Actions

Cell permeable fluorescent minor groove-binding probe for DNA. Binds to the minor groove of double-stranded DNA (preferentially to AT rich DNA), forming a stable complex which fluoresces approximately 20 times greater than DAPI alone.

  1. I have just received the product, but it did not include any instructions on how to use it. Could you please provide a protocol for using the dye solution (e.g., recommended incubation time for proper staining, etc.)?

    1. A protocol is not included with this product. This product is a ready?to?use, glycerol?based DAPI + antifade solution (1 µg/mL) supplied in a dropper vial for direct nuclear counterstaining on cells or tissues mounted on slides or coverslips. For general use, allow to warm to room temperature, gently swirl to mix, and protect from light. After fixation or immunofluorescence labeling and washes, apply enough solution to cover the specimen, coverslip, and incubate approximately 2–10 minutes at room temperature in the dark as a starting condition. If staining is too strong or bleeds into other channels, shorten incubation and optionally wash with PBS to reduce background before imaging. Image in the DAPI channel (DNA complex typically excitation ~360 nm, emission ~460 nm) and store the reagent at -20 °C, protected from light.
