Allgemeine Beschreibung

Das neue Triglycerid-Quantifizierungskit, MAK564, ist jetzt erhältlich! Triglyceride (TG) sind die Hauptbestandteile von pflanzlichen Ölen, tierischen Fetten, LDL und VLDL. Sie spielen eine wichtige Rolle für den Transport von Fettsäuren sowie als Energiequelle.[1][2] TG werden in Fettsäuren und Glycerin aufgespalten, die jeweils als Substrat für die Energie erzeugenden und Stoffwechselwege dienen.[3] Eine hohe Konzentration von Triglyceriden im Blut trägt zu Atherosklerose, Herzerkrankungen und Schlaganfällen sowie zu Pankreatitis bei.[4]

Eignung

Geeignet zur Verwendung mit Serum, Plasma und anderen biologischen Flüssigkeiten sowie in Gewebe- und Zellkulturproben.

Prinzip

Der Triglycerid-Quantifizierungs-Kit bietet einen empfindlichen, anwenderfreundlichen Assay zur Messung der TG-Konzentration in verschiedensten Proben. In diesem Assay werden Triglyceride in freie Fettsäuren und Glycerin aufgespalten. Das Glycerin wird dann oxidiert, was ein kolorimetrisches (570 nm) bzw. fluorometrisches (lex = 535 nm / lem = 587 nm) Produkt ergibt. Die Empfindlichkeit des Kits ist ausreichend, um 2 pmol–10 nmol (Bereich 2–10.000 mM) Triglyceride in unterschiedlichen Proben nachzuweisen. Das Kit erkennt außerdem Mono- und Diglyceride.

  1. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.   2. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  3. Can ethanol or isopropanol (IPA) be used instead of water for liver tissue sample preparation with the Triglyceride Quantification Colorimetric/Fluorometric Kit (MAK266)?

    1. Based on the guidelines, only water is an appropriate solvent for samples in this assay. Ethanol and isopropanol are not suitable solvents for this assay.

  4. Is liver tissue extracted using RIPA buffer and without NP-40 compatible with the MAK266 Triglyceride Quantification Colorimetric/Fluorometric Kit?

    1. It is recommended to use NP-40 or similar non-ionic detergents instead of RIPA, due to the presence of harsh detergents in RIPA.

    Terrible kit!

    The protocol is not well-written. I looked for several literatures who used this kit and found inconsistent data for control samples. Even the technical staffs of Sigma could not satisfy my questions and I strongly discourage other shoppers to use this.
