General description

Das neue Amylase-Testkit, MAK478, ist jetzt erhältlich! Amylase-Enzyme sind Glycosidasen, die Glucanverbindungen in Polysaccariden wie Stärke oder Glycogen spalten und so Polysaccarid-Moleküle freisetzen. α-Amylase ist in Tieren, Pflanzen und vielen Pilzen vorhanden. Bei Säugetieren ist α-Amylase die vorherrschende Form und eines der wichtigsten Enzyme für den Nahrungsverdau. Sowohl Aktivität als auch Stabilität von α-Amylase hängen von Calcium-Ionen ab. Beim Menschen ist der α-Amylasegehalt in den Sekreten von Pankreas und Speicheldrüsen am höchsten. Ein erhöhter Amylasegehalt in Blut und Urin ist auf Pankreaserkrankungen und Verletzungen der Speicheldrüsen zurückzuführen. Erhöhte Amylaseaktivität im Speichel ist außerdem ein Marker für erhöhte Aktivität des sympatischen Nervensystems.

Das Amylaseaktivität-Assaykit wird für die Bestimmung der Aktivität von Amylase verwendet.
Geeignet für die Quantifizierung der Amylaseaktivität in verschiedenen biologischen Proben.

Biochem/physiol Actions

Das Amylaseaktivität-Assaykit quantifiziert die Amylaseaktivität in verschiedenen biologischen Proben. Dieser Assay ist schnell, praktisch und empfindlich. In diesem Kit wird die Amylaseaktivität mithilfe eines gekoppelten enzymatischen Assays bestimmt, der ein kolorimetrisches (405 nm) Produkt ergibt, das proportional zum vorhandenen Substrat Ethyliden-pNP-G7 generiert wird, welches durch die Amylase gespalten wird. Eine Einheit ist die Menge an Amylase, die durch Aufspaltung von Ethyliden-pNP-G7 bei einer Temperatur von 25 °C in einer Minute 1,0 μmol p-Nitrophenol generiert.

  1. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  2. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.     1. This kit is not an ELISA assay, as there are no antibodies involved. The principle of this kit is that alpha-Amylase cleaves the substrate ethylidene-pNP-G7 to produce smaller fragments that are eventually modified by alpha-glucosidase. P-nitrophenol is then released, which leads to the color development that is monitored by absorbance at 405 nm spectrophotometrically.
