General description

Glutamat, eine der beiden sauren proteinogenen Aminosäuren, zählt auch zu den Schlüsselmolekülen für den Zellstoffwechsel. Die Synthese von α-KG aus Glutamat durch die Glutamatdehydrogenase ist eine anaplerotische Schlüsselreaktion im Citratzyklus. Beim Menschen spielt Glutamat eine wichtige Rolle sowohl beim Aminosäureabbau als auch bei der Entsorgung von überschüssigem oder als Abfall anfallendem Stickstoff. Im Säugetiernervensystem ist Glutamat der mengenmäßig häufigste exzitatorische Neurotransmitter. Es wird angenommen, dass es sowohl am Lernprozess und Erinnerungsvermögen beteiligt ist, als auch an Krankheiten wie der amyotrophen Lateralsklerose, Autismus, einigen Formen der geistigen Behinderung und der Alzheimer-Demenz. Glutamat kommt in vielen Nahrungsmitteln vor und wird in der Lebensmittelindustrie als Geschmackverstärker eingesetzt.

Geeignet zur Bestimmung der Glutamatkonzentration in verschiedenen Proben.
Das Glutamat-Assay-Kit wird zur Bestimmung der Konzentration von Glutamat im Liquor in einem Modell subarachnoidaler Hämorrhagie der Ratte verwendet. Es wird auch verwendet zur Konzentrationsbestimmung von zytosolischem und sezerniertem Glutamat im Liquor von Patienten mit amyotropher Lateralsklerose.

Biochem/physiol Actions

Die Glutamatkonzentration wird mit einem Enzymassay bestimmt, der proportional zur vorliegenden Glutamatmenge ein kolorimetrisches Produkt (450 nm) ergibt.

  1. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  2. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.     1. The Glutamate Assay Kit (MAK004) has a lower limit of detection of 40 µM and shows a linear standard curve up to 200 µM glutamate. One kit is designed to measure up to 100 samples.

  4. Why are cloudy white precipitates sometimes observed after reconstituting the enzyme mix in the Glutamate Assay Kit (MAK004)? Is there a way to reconstitute it?

    1. The presence of precipitates in the enzyme mix may be attributed to the reconstitution temperature. It is suggested to ensure complete equilibration of the Assay Buffer at room temperature before use. Reconstitute the Enzyme Mix according to the protocol using 220 µL Assay Buffer and keep it on ice during the assay. Gently vortexing is also an option. It is noted that the precipitates in the Enzyme Mix should not affect the assay, given that the mix is more concentrated than necessary. Care should be taken to avoid pipetting the precipitates into the wells. However, occasional precipitates in the Enzyme Mix should not have a detrimental impact.

  5. What is the reason for the lack of color change in the samples at 100 mM concentration, despite the expectation of a rapid or strong color change if the glutamate concentration is out of the standard range?

    1. The standard prepared at 10 nmole/well corresponds to 200 uM. To calculate this, you can use the formula M1V1 = M2V2 based on the preparation in the bulletin. The highest point standard provided in the kit is 200 uM. If the concentration exceeds the range of the standard curve, extrapolating the information for a decision is not appropriate. Dilution is necessary to ensure the concentration is within the standard curve for a correct determination of the glutamate concentration. A concentration of 100 mM is significantly out of range and may be causing the "Hook effect," leading to no color development. The "Hook effect" occurs due to the saturation of the reagents involved in the reaction for color production.
