General description

Chemische Struktur: Aminoglykosid
Kanamycinsulfat ist ein aus Streptomyces kanamyceticus gewonnenes Breitspektrum-Aminoglykosid-Antibiotikum. Es wird als Additiv in Kulturmedien für die Isolierung von Streptokokken der Gruppe D auf Kanamycin-Äsculin-Azid-Agar und für die Selektion von Neomycin-Phosphotransferase-haltigen transformierten Pflanzenzellen auf einem Kanamycin-Medium verwendet. Kanamycinsulfat kann auch als Selektionswirkstoff für Zellen, die mit dem Kanamycin-B-Resistenzgen transformiert wurden, verwendet werden. Für Zellkulturanwendungen sollte es in einer Menge von 100μg/ml zugegeben werden.

Kanamycinsulfat aus Streptomyces kanamyceticus wurde in Gewebekulturmedien verwendet.

Biochem/physiol Actions

Wirkungsweise: Das Produkt wirkt, indem es an die 70S-Ribosomen-Untereinheit bindet und so die Translokation und Auslösung von Fehlkodierungen hemmt.

Widerstandsweise: Aminoglycosid-modifizierende Enzyme (einschließlich Acetyltransferase, Phosphotransferase, Nukleotidyltransferase) können dieses Antibiotikum verändern und dessen Wechselwirkung mit Ribosomen verhindern.

Antimikrobielles Spektrum: Kanamycinsulfat ist wirksam gegen gramnegative und grampositive Bakterien und Mycoplasma.

Preparation Note

Kanamycinsulfat ist bei einer Konzentration von 50 mg/ml wasserlöslich und ergibt eine klare Lösung. In Alkohol, Aceton, Chloroform, Ether und Ethylacetat ist es praktisch unlöslich. Eine 1%-ige Lösung in Wasser hat einen pH-Wert von 6,5 bis 8,5. Sterile Lösungen können mittels Sterilfiltration durch einen 0,2μm Filter hergestellt werden.

Disclaimer

Lösungen sind bei 37 °C etwa 5 Tage stabil. Wässrige Stammlösungen können bei 2 - 8 °C langzeitig gelagert werden.

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.

  5. What is the difference between K1377 and K4000? K4000 is labeled as being free of animal-derived materials, but K1377 contains them?

    1. Both the K1377 and K4000 are animal component free. They are the same compound with the same properties, however the K1377 is also tested for use in mammalian and plant cell culture. See the links below for a sample Certificate of Analysis for each for comparison:
