Application

Wasserstoffperoxidlösung wird wie folgt verwendet:
  • zum Behandeln von Nasenepithelzellen für H2O2-Sensitivitätsassays
  • zum Behandeln der humanen unsterblichen Keratinozytenzelllinie (HaCaT) und der humanen Melanomzelllinie (GLL19) zum Einleiten der Seneszenz
  • zum Inkubieren von Hirnschnitten für die Bromdesoxyuridin (BrdU)/NeuN-Färbung
  • im Zellproliferationsassay zum Inkubieren von neuralen Stammzellen (NE-4C) für In-vitro-Experimente

Biochem/physiol Actions

Wasserstoffperoxid (H2O2) wird hauptsächlich für die Wundreinigung eingesetzt. Es kann Pathogene durch Oxidationsschübe und durch Erzeugen von lokalem Sauerstoff abtöten. H2O2 dient als Signalmolekül oder sekundärer Botenstoff. Es spielt auf zahlreiche Arten eine wichtige Rolle für die Genexpression. H2O2 ist an der Modulation von Entzündungen beteiligt.

  1. Hello, I am trying to analyse soil peroxidase activity but i have no results. I wonder if Hydrogen peroxide H1009 is normally the best product.

    1. Yes, this product is recommended. It is the hydrogen peroxide that is used in our Enzymatic Assay of Peroxidase, using Pyrogallol as the substrate. See: https://www.sigmaaldrich.com/US/en/technical-documents/protocol/protein-biology/enzyme-activity-assays/enzymatic-assay-of-peroxidase

      Please note that since this is a colorimetric assay, the color of the sample may interfere with the absorbance readings. So the soil sample may need to be decolorized before testing for enzymatic activity.

  2. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  3. How much volume do i need of 30% H2O2 if i want to add 0.3% of it to a 50 ml PBS solution?

    1. If 0.3% H2O2 (by volume) is to be achieved, it would be necessary to add 0.15 mL of H2O2, which would be 0.5 mL of the 30% solution. If the solution is to be exactly 0.3% H2O2, it would be best to only use 49.5 mL of PBS, to account for the added volume of H2O2 solution. If the 0.3% is to be m/v or v/v, the density would need to be used in order to convert.

  4. How much we need to add 30% H2O2 in 1mL of solution(water as solvent) to make100mM molarity?

    1. The molarity of this product is calculated to be 9.791 M based on a density of 1.11 g/mL, a formula weight of 34.01 g/mol, and a concentration of 30 w/w%. Here is the calculation:
      Molarity = Density x concentration x 1000 / formula weight = 1.11 x 0.3 x 1000 / 34.01 = 9.791 M.

      To make a 100 mM solution, use the Dilution Formula: C1V1 = C2V2 to find the volume V1 of the 30% H2O2 solution needed.
      C1 = 9.791 M (concentration of the stock solution)
      C2 = 0.1 M (desired concentration)
      V2 = 0.001 L + V1 (final volume, which is the sum of 1 mL of water and the volume of H2O2 added)

      Rearrange the formula to solve for V1:
      9.791 x V1 = 0.1 x (0.001 + V1)
      V1 = 0.1 x 0.001 / (9.791 - 0.1) = ~0.0000103 L = ~10.3 µL

      Therefore, approximately 10.3 µL of this product is required for a 100 mM H2O2 solution when mixed with 1 mL of water.

  5. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.   6. What stabilizer was added? I can't find any information in the DSD or spec sheet.

    1. This product contains Dipicolinic acid at 5 – 10ppm as a stabilizer.

  7. how can we determinate H2O2 concentration in case of 30% by titration , i found way to do this with sodium thiosulfate but they mentioned that is for 0.1% - 5 % con. - can i use it ?

    1. Using a procedure meant for lower concentrations of hydrogen peroxide may lead to less accurate results.   9. Hi, I would like to get information on this product (H2O2 Cat # H1009-5ML) for Lot numbers MKCL6061 and MKCJ1034 to know their expiration dates. Please help me get this information. Thanks.

    1. This product is not assigned an expiration date. It is assigned a recommended retest date of approximately one year after the quality release date. A recommended retest date is the time during which the product is expected to remain within established stability specifications, provided that it has been stored under defined conditions. After the Retest Date, product samples should be examined to ensure that the product is still in compliance with the established specifications.

  10. how much we need to add 30% H2O2 in 7ml of solution(water as solvent) to make 5mM molarity....could you please tell with calculation?

    1. The molarity of this product will change slightly from lot to lot, based on the final concentration. The concentration range is 29 - 32 %. The molarity can be calculated using the basic equation (density in g/L x final percentage)/molecular weight. The molarity of a 30 % solution of this product can be calculated by the following:
      (1110 g/L X 0.30) / 34.01 = 9.79 M.

      The volume of product required to prepare 7 mL of 0.005 M H2O2 would be 3.6 ul. For ease of pipetting, it may be more practical to prepare a larger volume. For example:
      Use 3.6 uL to prepare 7 mL at 5 mM
      Use 7.7 uL to prepare 15 mL at 5 mM
      Use 10.3 uL to prepare 20 mL at 5 mM

      Alternatively, an intermediate working solution can be prepared that would require further dilution.
      To prepare a 1 M stock from the 9.79 M concentrate, a dilution may be prepared using the M1V1 =M2V2 formula.
      In this case, 1.02 mL of the 9.79 M stock is diluted to 10 mL to achieve a 1 M concentration.
      From the 1 M solution dilute 0.035 mL (35 ul) to a final volume of 7 mL to achieve a 5 mM concentration.
