General description
Das Kit enthält alle Reagenzien, die für die Quantifizierung von DNA unter Verwendung des Fluoreszenzfarbstoffs Bisbenzimid (Hoechst 33258) erforderlich sind. Mit dem Kit sind hochempfindliche, schnelle und genaue Bestimmungen von doppelsträngiger DNA von 10 ng/mL bis 5 μg/mL möglich, auch in Gegenwart von RNA und Proteinen. Die Fluorometrie ist eine hochempfindliche und einfache Methode zur Quantifizierung von DNA. Der Fluoreszenzfarbstoff Bisbenzimid H 33258, der hauptsächlich an AT-Sequenzen in der Nebenrille der doppelsträngigen DNA (dsDNA) bindet, ist spezifisch für die Quantifizierung von DNA-Mengen im Nanogrammbereich (10 ng/mL bis 10 mg/mL). Bei Anregung mit 360 nm nimmt die Fluoreszenzemission des Farbstoffs bei 460 nm in Gegenwart von DNA deutlich zu.
Application
DNA-Quantifikationskit, Fluoreszenzassay wird zum Messen der Konzentration/des Gehalts von DNA in Folgendem eingesetzt:
- ventrikuläre Monozyten von neugeborenen Ratten (NRVM)
- humane mesenchymale Stammzellen (hMSC)/Proben zur Normalisierung der relativen ALP-Menge (alkalischen Phosphatase)
- embryonische Fibroblastenzelllinie der Maus (NIH3T3-Zellen)

The DNA standard (Product No. D4810) should be incubated at 50 °C for 15-30 minutes before determining the concentration spectrophotometrically. This will ensure complete dissolution of the genomic DNA.
