General description

Superoxide dismutase (SOD) is part of a family of metal ion-containing enzymes (SODs) that are found in most organisms, from bacteria to mammals. SODs catalyze the dismutation of superoxide (generated by aerobic respiration) to molecular oxygen and peroxide. Superoxide dismutase form the front line of defence against reactive oxygen species (ROS)-mediated injury and harbor anti-inflammatory activities. The Superoxide Dismutase (SOD) Activity Assay Kit provides a simple and sensitive procedure for measuring SOD enzymatic activity in various sample types. SOD activity is determined by measuring the decrease in superoxide anions (generated by the enzyme xanthine oxidase [XO]). Since superoxide anions interact with the provided WST dye, yielding color at 450 nm, the decrease in the color signal is proportional to SOD inhibition activity. The kit allows the determination of SOD activity in percent inhibition, or in units of activity using the provided SOD Enzyme as a standard. The provided SOD Enzyme can also be used to screen for SOD inhibitors.
The kit’s lowest limit of detection is 0.3 SOD units/mL. This kit can be used to measure SOD activity in biological samples such as serum, plasma and erythrocytes, tissue homogenates and cell lysates.

  2. Can this kit be used to measure bacterial superoxide dismutase activity?

    1. The Superoxide Dismutase, SOD, Activity Assay Kit (CS0009) can be used to measure SOD activity in bacteria. For cell and bacteria samples: Collect cell or bacteria into centrifuge tube, discard the supernatant after centrifugation, add 1 ml Assay buffer for 5×106 cell or bacteria, sonicate (with power 20%, sonication 3s, with an interval of 10s, repeat 30 times); centrifuged at 8000g 4 °C for 10 minutes, take the supernatant into a new centrifuge tube and keep it on ice for detection.

      Another option is to use CelLytic™ B Cell Lysis Reagent. For bacterial cell lysis, standard strength B7435 Product Description CelLytic™ B is used for the lysis of bacterial cells for the purification of recombinant and wild type proteins. CelLytic™ B contains 40 mM Tris-HCl, pH 8.0, and a proprietary, non-denaturing formulation of zwitterionic detergents. There is no need for special equipment to disrupt cells such as a sonicator or French press. This fast one-step method obtains higher yields than sonication, lysozyme treatment, or other commercially available lysis solutions.

  3. How many cells are suggested for each sample to get a good reading?

    1. Unfortunately, no internal information is available regarding the number of cells to use with this product, as the number of cells needed can vary depending on the cell type. Conducting a pilot experiment to optimize the cell numbers for the assay is recommended. A good starting point is 2 x 10^6 cells, but this may need to be adjusted based on cell type.

  8. How much serum sample volume needed for this assay?

    1. All assays (samples, standards, controls and blank) require 20 μL of sample for each reaction (well). Therefore, bring the sample volume to 20 μL. When required, samples should be diluted in Dilution Buffer.
