Das Enzym wird verwendet, um die Folgen der Induzierung akuter, langfristiger Veränderungen von Chondroitinsulfat-Proteoglykanen in den Hirnzellen adulter Ratten zu untersuchen.[1] Glycosaminoglykane werden mit Chondroitinase ABC für die Chondroitin-/Dermatansulfat-Quantifizierung in Keratozyten-Zellkulturen verdaut, die aus frischem Hornhautstroma von Rindern gewonnen werden.[2]
Biochem/physiol Actions
Chondroitinase ABC katalysiert den eliminativen Abbau von Polysacchariden, die (1-4)-β-D-Hexosaminyl und (1-3)-β-D-Glucuronosyl enthalten oder (1-3)-α-L-Iduronosyl-Bindungen an Disaccharide, die 4-Deoxy-β-D-gluc-4-enuronosyl-Gruppen enthalten. Es wirkt auf Chondroitin-4-Sulfat, Chondroitin-6-Sulfat und Dermatansulfat ein sowie langsam auf Hyaluronat. Das Molekulargewicht beträgt ungefähr 120 kDa, wobei 2 unterschiedliche Untereinheiten einer Molekularmasse von 86 kDa und 32 kDa vorliegen. Der optimale pH-Wert beträgt 8,0 bei Chondroitinsulfat und 6,8 bei Hyaluronsäure; die optimale Temperatur liegt bei 37 °C. 1 mM Zn2+ wirkt hemmend, während 0,05 M Acetat das Enzym aktivieren.

Could you provide specific data regarding the pH-dependent activity of Chondroitinase ABC (Product No. C3637)? I am particularly interested in how the enzymatic activity changes between pH 7.0 and pH 8.0.

This product shows maximal enzymatic activity on chondroitin sulfate at approximately pH 8.0, based on assay conditions at 37 °C. At pH 7.0, the enzymatic activity rate is significantly lower. The pH-activity profile indicates activity peaks at pH 8.0 and decreases to less than 40% when the pH falls below 7.0, indicating a steep decline. For hyaluronan, the optimal pH is approximately 6.8. This product should be reconstituted in a 0.01% BSA aqueous solution. Subsequent dilutions can be made into a buffer containing 50 mM Trizma® HCl, pH 8.0, with 60 mM sodium acetate and 0.02% BSA Prepare solutions just prior to use.
