General description
Accutase®-Lösung (in Dulbeccos Phosphatgepufferter Kochsalzlösung (DPBS) ohne Ca++ oder Mg++) enthält proteolytische und kollagenolytische Enzyme für die Zellablösung. Sie eignet sich zum Ablösen von Zellen von Standardbehältnissen aus Kunststoff für die Gewebekultur und von adhäsionsbeschichteten Kunststoffbehältnissen in Routineanwendungen. Accutase wird bei der Zellablösung gegenüber Trypsin bevorzugt, da sie für die Zellen schonender ist. Daher wird Accutase häufig für die Behandlung empfindlicher Zellen oder für die Analyse von Oberflächenmarkern verwendet.
Application
Nachweislich effektiv bei der Ablösung von primären Fibroblasten, Endothelzellen, Neuronen, Tumorzelllinien und Insektenzellen. Ausgezeichnete Leistung bei der Ablösung von Zellen zur Analyse von Zelloberflächenmarkern, Virenwachstumsassays, Durchflusszytometrie und beim Bioreaktor-Scaleup. Enthält keine Säuger- oder Bakterienprodukte.
Accutase® Lösung wird zur Dissoziation oder zum Aufschluss nicht-adhärenter, im Aggregat wachsender Zellen wie Hirntumorzellen, induzierte pluripotente Stammzellen (IPSC) und Glioblastomzellen[1] in einzelne Zellen verwendet.

Gebrauchsfertige Lösung mit einem proprietären Gemisch aus proteolytischen und collagenolytischen Enzymen zur Dissoziation von Zellen von haftbeschichteten und Standard-Kunststoffmaterialien.
Preparation Note
Dieses Produkt enthält Phenolrot. Das Produkt wird auf Trockeneis versandt; in der Verpackung kann ein bedeutender CO2-Aufbau stattfinden. Dieses CO2 kann in die Lösung übergehen und den pH-Wert leicht senken (auf etwa 6,8), was einen gelborangfarbenen Ton zur Folge hat. Die Lösung sollte dennoch verwendbar sein. Es kann auch Natriumhydroxid zugegeben werden, um den pH-Wert einzustellen. Eine vollständig gefrorene Accutase-Flasche weist eine gelbe Farbe auf, die beim Auftauen in einen orangefarbenen Ton umschlägt. Nach dem Auspacken kann das Produkt wieder eingefroren oder zur späteren Verwendung direkt in den Kühlschrank gegeben werden. Das Produkt nicht in einem warmen Wasserbad auftauen. Accutase ist empfindlich gegenüber Temperaturen über 37 °C und wird nach 45 Minuten bei 37 °C inaktiv. Empfohlen wird ein Auftauen über Nacht im Kühlschrank oder für 1 bis 1,5 Stunden in einer Wanne mit kaltem Leitungswasser. Die Flasche mit Accutase nach dem Auftauen gut schütteln, damit die Inhaltsstoffe gleichmäßig in der Lösung verteilt werden. Diese Produkte müssen vor der Verwendung nicht erwärmt werden. Nach der Entnahme aus dem Kühlschrank sofort verwenden und nach Gebrauch wieder in den Kühlschrank zurückstellen.
Zubereitet in Dulbeccos PBS (0,2 g/l KCl, 0,2 g/l KH2PO4, 8 g/l NaCl und 1,15 g/l Na2HPO4) mit 0,5 mM EDTA•4Na und 3 mg/l Phenolrot. Informationen zur Produktanwendung finden Sie online im Produktdatenblatt.

Yes, this product is stable after undergoing several freeze/thaw cycles. The thawed product may be aliquoted and stored at –20 °C or stored for up to 2 months at 4°C.

On the Innovative cell technologies website it says that accumax is the same as accutase but with 3x the amount of enzymes. Why did the technician here answer that it contains DNAse?

Thank you for calling this to our attention. The manufacturer, Innovative Cell Technologies, has been contacted regarding this matter. The manufacturer has confirmed that both Accutase and Accumax products DO contain a small amounts of DNAse. The concentration is not measured, but it is present in the formulation. In addition, both products also contain 0.5 mM EDTA.

How can I inactivate Accutase® with fetal bovine serum?

Due to its gentle nature Accutase® cell detachment solution usually does not need to be neutralized following cell passaging. Accutase® is also heat inactivated after 45 minutes at 37°C. If this method of neutralization does not work for your cells, or if you want to neutralize Accutase® in a shorter period of time, the addition of 10% FBS containing media will accomplish this task. Experiments have shown that diluting a sample of Accutase® with 10% with fetal bovine serum achieved an environment similar to that found in cell culture. Measurements of the enzyme activity in this diluted sample showed decreased activity over time, falling below that of the required level for cell detachment (>175 units/mL). After about 6 minutes of serum interaction, the enzyme activity of Accutase® was well below this level, demonstrating effective inhibition by FBS.

What is the appropriate cell dissociating product to be used for ES cells that will be injected into blastocysts?

Accutase®, product A6964, or Accumax™ (Accutase® with DNase), product A7089, will work well for dissociation of ES cells. The Accutase® product has protease and dispase activity to dissociate the cells.
