Das Cell Counting Kit-8 (CCK-8) basiert auf WST-8 (2-(2-methoxy-4-nitrophenyl)-3-(4-nitrophenyl)-5-(2,4-disulfophenyl)-2H-tetrazolium, Mononatriumsalz), das in Gegenwart des Elektronenträgers 1-Methoxy PMS in einen wasserlöslichen Formazanfarbstoff umgesetzt wird. Die CCK-8-Lösung wird den Zellen direkt zugegeben. Ein Vormischen der Komponenten ist nicht erforderlich. WST-8 wird durch zelluläre Dehydrogenasen in ein orangefarbenes Formazanprodukt umgesetzt, das sich im Zellkulturmedium löst. Die Menge des gebildeten Formazans ist direkt proportional zur Zahl der lebenden Zellen. Da die CCK-8-Lösung sehr stabil und wenig zytotoxisch ist, ist eine längere Inkubation von 24 bis 48 Stunden möglich.

100 Test means 100 wells, so only one 96-well plate?

The reagents provided in kit 96992 are sufficient for 100 tests. A 96 well microplate is not included with the kit. The customer must provide the 96 well plate. Although a solid 96 well plate can be used, a 96 well microplate with removable strips will allow for the most efficient use of the kit.

Is this kit suitable for use with HEK293 or HEK293f cells?
