General description
Ampicillin-Natriumsalz gehört zur β-Lactam-Familie mit breitem Wirkungsspektrum und ist ein semisynthetisches Penicillin-Derivat, das als Breitspektrum-Antibiotikum eingesetzt wird. Es handelt sich um ein β-Lactam-Antibiotikum, das die Zellwandsynthese von Bakterien durch Bindung an Penicillin-bindende Proteine (PBPs) und dadurch die Synthese von Peptidoglykan hemmt, welches einen entscheidender Bestandteil der bakteriellen Zellwand ausmacht. Ampicillin-Natriumsalz wirkt aktiv gegen eine Vielzahl grampositiver und gramnegativer Bakterien, einschließlich E. coli, β-Lactam-sensitive VRE, Vancomycin-resistente Enterokokken, Staphylococcus aureus und Streptococcus pneumoniae. Es wird häufig in Forschungslaboren eingesetzt, um Antibiotikaresistenzen und Penetrationsfähigkeiten sowie die Synergie zwischen verschiedenen Antibiotika und die Auswahl und Erhaltung rekombinanter Plasmide bei E. coli zu untersuchen.

Wirkungsweise der Resistenz: Die Verabreichung zusammen mit β-Lactamase spaltet den β-Lactam-Ring von Ampicillin und deaktiviert ihn.
Antimikrobielles Spektrum: Enthält sowohl grampositive (Benzylpenicillin-ähnliche) wie auch gramnegative (Tetracyclin- und Chloramphenicol-ähnliche) Bakterien.

Ampicillin ist laut Berichten schwer in Wasser löslich, so gut wie unlöslich in Alkohol, Chloroform, Ether und fetten Ölen, aber löslich in verdünnten Säuren oder Basen. Die Lösung sollte nicht autoklaviert werden; eine Stammlösung sollte sterilfiltriert und eingefroren gelagert werden, wodurch sie monatelang haltbar ist.

Zusätzliche Studien haben gezeigt, dass die Stabilität von Ampicillin in Lösung vom pH-Wert, der Temperatur und dem verwendeten Puffer abhängt. Seine Aktivität geht schnell verloren, wenn es bei einem pH-Wert von über 7 gelagert wird. Als optimale Lagerungsbedingungen werden 2–8 °C und ein pH-Wert von 3,8–5 empfohlen, bei denen die Aktivität eine Woche lang bei über 90 % erhalten wurde.

How do I use Ampicllin in agar plates?

For plates, add ampicillin to autoclaved media cooled to 48°C, to a final concentration of 35-50 μg/mL. Pour the plates, and allow them to cool completely. Plates may be stored at 2-8°C for up to only 1-2 weeks before use.
