Application

Sudan Black B has been used in brain BLAQ (Block Lipids and Aldehyde Quench) technique and is used in lipid detection.
Zertifiziert für die Fettfärbung nach der Churukian Variante des Lillie und Asburn-Verfahrens ("supersaturated isopropanol method"). Sudan III ersetzt Ölrot O in dem Verfahren. Burdon-Färbung für bakterielle Lipide.

Biochem/physiol Actions

Sudan Black B has been used to stain phospholipids and intracellular lipids. Staining pattern is almost similar to myeloperoxidase reaction, with positive staining of granulocytic cells, eosinophils and weak monocytic staining.It doesn′t stain lymphocytes. Sudan Black B can be used to stain older blood or bone marrow smears, but the stain slightly fades over time. It is considered to be a most sensitive and specific reagent for lipid staining. Sudan Black B stains phospholipids gray and neutral fats blue-black.

    1. Biological dyes do not ship with protocols for use. There are published procedures for Sudan Black B for cytology that provide a formulation using Phosphate-buffered Phenol. The Atlas of CytoChemistry and Immunohistochemistry of Hematologic Neoplasms by Sun, Li and Yam, ASCP Press, 1985 gives the following formulation:
      Dissolve 32 grams of crystal Phenol in 60 ml of absolute Ethanol. Once dissolved, prepare the Phosphate buffer solution. Add 0.6 grams Sodium Phosphate, then add distilled water until the volume reaches 200ml.
      The instructions then state to prepare the working solution by adding 60 ml of the alcoholic Sudan Black B stock solution to 40 ml of the Phenol Phosphate buffer.

  2. Can Sudan black used for Immunofluorescence specifically for paraffinized eyeball sectioning?

    1. Sudan Black B is not a fluorescent dye. Sudan Black B has been used to block autofluorescence. The following information is available from the website. Although the reference below is specific to brain tissue, the expectation is that Sudan Black B should be able to be used for a variety of different tissue types. There is no specific information available for paraffin sections of eyeballs.

      Sudan Black B treatment reduces autofluorescence and improves the resolution of in situ hybridization-specific fluorescent signals of brain sections.

      ABSTRACT
      Interference by autofluorescence is one of the major concerns of immunofluorescence analysis of in situ hybridization-based diagnostic assays. We present a useful technique that reduces autofluorescent background without affecting the tissue integrity or direct immunofluorescence signals in brain sections. Using six different protocols, such as ammonia/ethanol, Sudan Black B (SBB) in 70% ethanol, photobleaching with UV light and different combinations of them in both formalin-fixed paraffin-embedded and frozen human brain tissue sections, we have found that tissue treatment of SBB in a concentration of 0.1% in 70% ethanol is the best approach to reduce/eliminate tissue autofluorescence and background, while preserving the specific fluorescence hybridization signals. This strategy is a feasible, non-time consuming method that provides a reasonable compromise between total reduction of the tissue autofluorescence and maintenance of specific fluorescent labels.

  3. How to dissolve Sudan Black B for staining tissue?

    1. Sudan Black B can be dissolved in a variety of different solvents depending upon the intended use. Dissolve 0.7 grams of Sudan Black B in 100 ml of propylene glycol. The solution should be heated to 100° C for a few minutes, stirred constantly, and then filtered for optimal results.
