Beschreibung

General description

Ein hochpotenter, zellpermeabler, reversibler und selektiver Inhibitor von Rho-assoziierten Proteinkinasen (K_i = 140 nM für p160^ROCK). Hemmt auch ROCK-II mit gleicher Potenz. Die Hemmung ist in Bezug auf ATP kompetitiv. Weist eine um das 10- bis 50-fache niedrigere Affinität gegenüber PKC_ε auf als p160^ROCK. Hat keine Auswirkungen auf die Aktivität von p21-aktivierter Proteinkinase (PAK), selbst bei höheren Konzentrationen (~100 µM). Fungiert auch als potenter Inhibitor einer Agonist-induzierten Ca²⁺ Sensibilisierung der Myosin-Phosphorylierung und der Glattmuskelkontraktion. Verhindert eine Apoptose und verbessert die Überlebens- und Klonierungseffizienz von dissoziierten hES-Zellen ohne Beeinflussung ihrer Pluripotenz.
