Beschreibung

General description

Ein zellpermeabler, potenter, selektiver, konkurrierender und reversibler Inhibitor von CD45 (IC₅₀ = 200 nM bei Verwenung von pNPP als Substrate, 3,8 µ;M für CD45 lck, >;30 µ;M für PTP1B lck) und weist anti-proliferative Eigenschaften auf. Berichten zufolge ist es ein starker Blocker der T-Zellrezeptor-vermittelten Proliferation (IC₅₀ = 100 nM). Ist weniger zytotoxisch (CC₅₀ = 3,5 µ;M) als andere 1,2-Naphthalendion-Analoge. Die Gruppe der Phenanthrenedione wirkt nicht als Thiol-oxidierender Wirkstoff. Einer der stärksten Hemmer der PTEN (phosphatase and tensin homolog deleted on chromosome 10), der den PIP3-Signalweg in verschiedenen Zellen stärkt und die Wirkung der Granulozytentransfusion in murinen Modellen der neutropenieassoziierten Pneumonie verbessert.
