General description
Eine symmetrische Bicyclam-Verbindung, die die CXCL12-Bindung (SDF1) an CXCR4 antagonisiert (IC50 = 108 und 245 nM für Ratten- bzw. menschliches CXCR4) und den SDF1-induzierte Ca2+-Fluss (um 100 % bei 100 ng/ml in SUP-T1- und THP-1-Kulturen) hemmt; ebenso hemmt sie eine CXCR4-, jedoch nicht CCR5-vermittelte HIV-Infektion (IC50 ≤200 nM) und verstärkt zugleich die Bindung von SDF1 an CXCR7 (um ~60 % bei 1 mM mit CXCR7-exprimierenden HEK293T-Zellen) und die SDF1-induzierte β-Arrestin-Rekrutierung an CXCR7 (EC50 = 6,48 und 11,8 nM bei Vorliegen bzw. Abwesenheit von 10 µM AMD3100). Eine Verabreichung von AMD3100 mittels intravenöser Infusion führt gemäß Berichten zur Mobilisierung hämatopoietischer Stammzellen in Menschen, Hunden und Mäusen in vivo. Weist keine hemmende Wirkung gegen den chemokin-induzierten Signalweg über CXCR1/2/3 oder CCR1/2/3/4/5/6/7/8/9 auf. Auch als 50-mM-Lösung in H2O (Katalog-Nr. 239825) erhältlich.
